林麝IGF1基因序列分析及多态性与产香量的关系研究

摘要 目的研究林麝IGF1基因多态性与产香量关系。方法利用PCR对不同产香量林麝IGF1基因进行扩增测序,分析林麝IGF1基因序列与近源物种的差异并寻找与林麝产香有关的SNP位点。结果林麝IGF1基因第三外显子与牛IGF1基因第三外显子序列存在四个碱基位点不一致,分别为全长CDS第246位G>C,第333位G>A,第360位C>T,369位T>C,碱基位点并不改变氨基酸的变化,但是能够影响mRNA对tRNA的亲和力和RNA二级结构,从而影响蛋白的正确折叠和表达情况;同时林麝IGF1基因第三内含子存在一个C/G的SNP位点,该位点C基因频率占70%,G基因频率占30%,不符合哈代温伯格平衡,说明该位点存在选择力,而在高产麝香组和低产麝香组中基因型分布差异不显著(P>0.05)。结论该研究可为生产实践中提高林麝麝香产量的分子选育提供参考。 Objective In order to explore the relationship between the polymorphism of IGF1 gene and yield of Moschus berezovskii.Methods The IGF1 gene amplification of M.berezovskii were performed by PCR,and the relevant fragments and difference among similar species were analyzed while the SNP sites related to musk production were screened.Results The results showed that the third exon of M.berezovskii IGF1 gene was inconsistent with the bovine genome sequence with four base sites including the 246th G>C,333rd G>A,360th C>T,369th T>C of all CDS sequence.The base sites did not change the amino acid changes,but could affect the affinity of mRNA to amino acid and the secondary structure of RNA,thus affecting the correct folding and expression of IGF1 protein.Meanwhile,there is a C/G SNP locus in the third intron of IGF1 gene of forest musk deer,with C gene frequency accounting for 70%and G gene frequency accounting for 30%,which is not in consistent with Hardy Weinberg equilibrium,indicating that this locus has selectivity.There was no significant difference in genotype distribution between high-yielding musk group and low-yielding musk group(P>0.05).Conclusion In the present,all the results will provide reference for improving musk yield of M.berezovskii in practice via molecular breeding.
