响应面法优化花生根白藜芦醇酶提工艺及不同品种含量分析

Response Surface Methodology to Optimize the Enzymatic Extraction Process of Resveratrol from Peanut Roots and the Content Analysis of Different Varieties

摘要 采用响应面优化酶法提取花生根中白藜芦醇(Resveratrol,Res)四种异构体的工艺条件,探究酶的种类、添加量、温度、时间、pH对Res提取量的影响,并利用超高效液相色谱(UPLC)分析19种不同品种花生根中Res和Res苷总含量。结果表明,最佳提取工艺:β-葡萄糖苷酶的添加量0.32 mg、温度70.89℃、pH=5.37、酶解时间121.71 min,在此条件下,Res提取量可以达到3719.7 mg/kg。在不同品种花生根中,花育系列花生根中Res和Res苷总含量高于其它品种,且花育967花生根中总含量最高为4137.18±19.56 mg/kg。该提取工艺为花生根中Res和Res苷的高效提取利用提供了技术基础。 The process conditions of the four isomers of resveratrol from peanut roots were extracted by response surface optimization enzyme method to explore the effects of enzyme species,addition amounts,temperature,time and pH on the extraction volume of Res,and analyze the total content of Res and Res glycosides in the roots of19 different varieties using ultra-high performance liquid chromatography(UPLC).The results showed that the best extraction process:the addition amount ofβ-glucosidase was 0.32 mg,the temperature was 70.89℃,the pH was5.37,and the enzymatic hydrolysis time was 121.71 min.Under these conditions,the extraction amount of Res could reach 3719.7 mg/kg.Among different varieties of peanut roots,the total content of Res and Res glycosides in the roots of Huayu series peanuts was higher than that of other varieties,and the total content of Huayu 967 peanut roots was the highest at 4137.18±19.56 mg/kg.The extraction process provides a technical basis for the efficient extraction and utilization of Res and Res glycosides from peanut roots.
