2种新疆小麦根际短体线虫的鉴定

摘要 新疆是我国西北重要的小麦优势产区,短体线虫是危害小麦的主要病原生物之一。采集了来自新疆不同地区的小麦根际土样,运用形态结合分子特征分析方法鉴定了其中的短体线虫种类。通过显微观察,形态测量,比较不同地区线虫的形态特征。同时,扩增了不同地区种群的rDNA 18S与28S D2-D3区基因片段序列,并用贝叶斯法构建系统进化树。结果表明,新疆小麦根际短体线虫主要为落选短体线虫P.neglectus和桑尼短体线虫P.thornei两种,其中落选短体线虫为优势种群,桑尼短体线虫为新疆植物线虫新纪录种。研究结果对于指导新疆小麦线虫病害防治、保障粮食生产安全具有重要意义。 Nematode of Pratylenchus is one of the main pathogens that harm wheat.In this study,the rhizosphere soil samples of wheat were collected from different areas in Xinjiang,an important wheat-producing area in northwestern China.Species of Pratylenchus were identified by morphological and molecular systematic analysis methods.Through microscopic observation and morphological measurement,the morphological characteristics of nematodes in different areas were compared.In the meanwhile,the rDNA 18 S and DNA28 S D2-D3 region gene fragments in different area populations were amplified,and the phylogenetic trees were constructed using Bayesian method.The results showed that the two species of Pratylenchus separated from wheat rhizosphere soil of wheat were P.neglectus and P.thornei,and among them,P.neglectus was the dominant population,P.thornei was a new record species in Xinjiang.The research results were of great significance for guiding the control of wheat nematode diseases and ensuring food production safety in Xinjiang.
