吐鲁番斗鸡与新罗曼鸡MAOA基因外显子区遗传多态分析

摘要 旨在把分子标记辅助选择应用于吐鲁番斗鸡攻击行为的选育。试验参照GenBank的鸡单胺氧化酶A(MAOA)基因DNA序列,利用软件Primer 5.0设计了10对引物,采用聚合酶链式反应-单链构象多态(PCR-SSCP)方法,对103只吐鲁番斗鸡和73只新罗曼鸡MAOA基因的10个外显子进行多态性检测分析。结果表明:MAOA基因的MAOA1、MAOA2、MAOA5、MAOA7扩增的片段检测到多态,其中引物MAOA1、MAOA5、MAOA7扩增的位点NC006088.5(25424-25662)、NC006088.5(34980-35257)、NC006088.5(38401-38613)在吐鲁番斗鸡和新罗曼鸡上均检测到多态,MAOA2引物扩增位点NC006088.5 (30260-30535)只在吐鲁番斗鸡上检测到多态。对检测到的多态性位点进行测序,经序列对比这些突变均为同义突变。经X2适合性检验表明,研究中出现多态性的基因位点的基因频率和基因型频率都符合Hardy-weinberg平衡状态(P>0.05)。用X2独立性检验表明,研究中出现多态性的基因位点的基因型的构成,与品种间有极显著性差异(P<0.01),说明MAOA基因与鸡的攻击行为有一定的联系。 In order to identify the molecular markers related to the aggressive behavior of Turpan fighting chickens, the molecular marker-assisted selection was applied to the breeding of Turpan fighting chickens to identify their behavioral traits. The primers were designed according to the chicken MAOA sequence using the software Primer 5. The polymorphism of 10 exons of the MAOA gene was analyzed by PCR-SSCP. The results showed that the fragments amplified by primers MAOA1, MAOA2, MAOA5, and MAOA7 showed polymorphism. The fragments NC006088.5(25424-25662), NC006088.5(34980-35257), and NC006088.5(38401-38613) amplified by primer MAOA1, MAOA5, and MAOA7 showed polymorphism in both Turpan fighting chickens and New Roman chickens, and the fragment NC006088.5(30260-30535) amplified by primer MAOA2 showed polymorphism only in the Turpan fighting chickens. The results of the X2 test showed that the frequencies of the MAOA gene reached the Hardy-weinberg equilibrium in the two breeds of chickens(P>0.05). The distribution and constitution of the genotype at these polymorphic sites of the MAOA gene showed were of significant differences between the Turpan fighting chickens and New Roman chickens(P<0.01), which implied that the MAOA gene had a relationship with the aggresive behavior of chickens.
