刺梨水溶性多糖提取工艺优化及其抗肿瘤活性评价 被引量:7

Optimization of the Water-Soluble Polysaccharide Extraction from Rosa Roxbunghii Tratt and Its Antitumor Activity

摘要 以刺梨多糖得率为考察指标,对热水浸提刺梨多糖工艺进行优化,并通过建立S180实体瘤模型对最优条件下提取的刺梨多糖进行抗肿瘤活性评价。结果表明:热水浸提刺梨多糖的最优工艺条件为液料比20 mL/g,浸提时间2.6 h,浸提温度90℃,浸提3次。在最优工艺条件下,刺梨多糖得率为(3.37±0.03)%(n=3)。在灌胃剂量200 mg/kg时,热水浸提刺梨多糖对S_(180)肿瘤小鼠的抑瘤率为(50.61±1.06)%,比超声辅助提取刺梨多糖活性更强,并能显著提高肿瘤小鼠的白细胞数量、胸腺指数和脾脏指数,说明在此工艺条件下提取的刺梨多糖,具有一定的提升肿瘤小鼠免疫能力和抗肿瘤作用,并具有成为功能性食品添加配料或天然抗肿瘤药物的潜力。 Taking the yield of Rosa roxburghii Tratt polysaccharide as the index,the hot water extraction process of Rosa roxburghii Tratt polysaccharide was optimized,and the anti-tumor activity of Rosa roxburghii Tratt polysaccharide was evaluated by establishing S180 solid tumor model.The results showed that the optimum extraction conditions for RRTP as follows:liquid to solid 20 mL/g,extraction time 2.6 h,extraction temperature 90℃,3 times of extraction.The yield of RRTP was(3.37±0.03)%on average(n=3)under the optimum processing conditions.Besides,antitumor activity analysis showed that RRTP has obvious antitumor activity and can significantly increase leukocyte count,thymus index and spleen index in S180 solid tumor mice.And the result indicates the RRTP extracted by hot water has a tumor inhibitory rate of(50.61±1.06)%in S_(180)solid tumor mice,stronger than ultrasound-assisted extraction of RRTP at the gastric irrigation dose at 200 mg/kg.It is concluded that RRTP could be used as a potential functional food supplements or natural substances of antitumor drugs.
