摘要
目的:构建一种兔类风湿性关节炎(rheumatoid arthritis,RA)动物模型并对模型效果进行评价,为RA的发病机制与治疗研究提供可靠的造模方法。方法:购置6月龄雄性新西兰兔26只,随机分为两组,共52例膝关节,分为对照组(4例)与模型组(48例)。以卵清蛋白(ovalbumin,OVA)法建立兔RA模型,对模型组行2次基础致敏与4次局部致敏,对照组不做任何处理。观察并记录兔膝关节表面温度、膝关节周径、体重等体征指标;使用彩色多普勒超声测量膝关节滑膜厚度、积液深度、滑膜血流分级,采集滑膜并制作病理切片,进行病理评分与病理分级,记录上述临床指标。对比上述体征指标与临床指标,评价模型建立情况。结果:建模操作后,模型组与对照组在膝关节表面温度、体重、膝关节滑膜厚度、膝关节积液深度、滑膜血流分级、病理评分、病理分级等指标差异有统计学意义(P<0.05),模型成功率为91.7%(44/48)。结论:通过此建模方法,可以构建成功率较高的兔类风湿性关节炎模型。
Objective To establish an animal model of rheumatoid arthritis(RA)in rabbits and evaluate the effect of the model,and to provide a reliable modeling method for the study of pathogenesis and treatment of RA.Methods Twenty-six male New Zealand rabbits aged 6 months were purchased and randomly divided into 2 groups,including 52 knee joints,which were divided into control group(4 cases)and model group(48 cases).Rabbit RA model was established by ovalbumin method.The model group was sensitized twice for basic sensitization and four times for local sensitization,while the control group was not treated with any treatment.The knee surface temperature,knee circumference,body weight and other physical signs of rabbits were observed and recorded.Color Doppler ultrasonography was used to synovial thickness,effusion depth and synovial blood flow grade of knee joint;Synovial specimens were collected and pathological sections were made.The results of each index were compared to evaluate the establishment of the model.Results After modeling operation,the differences in knee surface temperature,body weight,knee synovial thickness,knee fluid depth,synovial blood flow grade,pathological score and pathological grade between the model group and the control group were statistically significant(P﹤0.05),and the success rate of the model was 91.7%(44/48).Conclusion Through this modeling method,a rabbit model of rheumatoid arthritis with a high success rate can be constructed.
