α-平滑肌肌动蛋白、乏氧诱导因子-1α在肺癌中的表达及对放化疗疗效的影响

Expression ofα-SMA and HIF-1αin lung cancer and its effects on the efficacy of chemoradiotherapy

目的探讨α-平滑肌肌动蛋白(α-SMA)和乏氧诱导因子-1α(HIF-1α)在肺癌中的表达及对放化疗疗效的影响,以期为治疗肺癌提供新的靶标。方法选取66例肺癌患者为肺癌组,同期健康体检者66例为对照组,采用荧光定量PCR测定α-SMA和HIF-1α的mRNA表达水平;采用免疫组化法测定α-SMA和HIF-1α的蛋白表达水平;观察肺癌患者的疗效、毒副反应和2年生存率。结果肺癌组织α-SMA mRNA和HIF-1αmRNA相对表达量均高于癌旁组织和正常组织,差异均有统计学意义(P<0.05)。肺癌患者中α-SMA阳性表达为47例(71.21%),HIF-1α阳性表达为51例(77.27%)。肺癌患者α-SMA阳性组和HIF-1α阳性组中的放化疗有效率、2年生存率均分别低于α-SMA阴性组和HIF-1α阴性组,差异均有统计学意义(P<0.05)。结论肺癌患者中α-SMA、HIF-1α均呈现高阳性率,α-SMA、HIF-1α阳性患者放化疗有效率和2年生存率下降,α-SMA和HIF-1α阳性表达的相关分子机制还需进一步探究。

Objective To explore the expression ofα-SMA and HIF-1αin lung cancer and their effects on the efficacy of chemoradiotherapy,so as to providing new targets for the treatment of lung cancer.Methods Sixty-six lung cancer patients were selected as the lung cancer group,and 66 healthy subjects were selected as the control group.The mRNA expression levels ofα-SMA and HIF-1αwere detected using fluorescent quantitative PCR.The protein expression levels ofα-SMA and HIF-1αwere compared wih immunohistochemistry.The efficacy,toxicity and 2-year survival rate of lung cancer patients were observed.Results The relative expressions ofα-SMA mRNA and HIF-1αmRNA in lung cancer tissues were higher than those in adjacent tissues and normal tissues,and the differences were statistically significant(P<0.05).There were 47 cases(71.21%)with positive expression ofα-SMA,and 51 cases(77.27%)with positive expression of HIF-1αin lung cancer patients.The efficiency of chemoradiotherapyand 2-year survival rate in theα-SMA-positive group and HIF-1α-positive group of lung cancer patients were lower than those in theα-SMA-negative group and HIF-1α-negative group,respectively(P<0.05).Conclusions The positive rates ofα-SMA and HIF-1αwere high in lung cancer patients.The efficiency of chemoradiotherapy and the 2-year survival rate ofα-SMA-positive and HIF-1α-positive patients were decreased.The related molecular mechanisms ofα-SMA and HIF-1αpositive expression need to be further explored.
