LncRNA ZFAS1/miR-150-5p轴表达与胃癌根治术患者病理特征及预后的相关性

Correlation between the expression of lncRNA ZFAS1/miR-150-5p axis and the pathological characteristics,prognosis of patients undergoing radical gastrectomy

目的研究LncRNA ZFAS1/miR-150-5p轴表达与胃癌根治术患者病理特征及预后的相关性。方法选择2015年在冀中能源峰峰集团有限公司总医院进行胃癌根治术的患者,收集胃癌组织及癌旁组织,检测lncRNA ZFAS1、miR-150-5p的表达水平并分析其相关性,随访胃癌患者的总生存期,采用Kaplan-Meier曲线分析不同lncRNA ZFAS1、miR-150-5p表达水平的胃癌患者总生存期的差异。结果与癌旁组织比较,胃癌组织中lncRNA ZFAS1的表达水平明显增加、miR-150-5p的表达水平明显降低(P<0.05),且胃癌组织中lncRNA ZFAS1的表达水平与miR-150-5p的表达水平呈负相关(r=-0.392,P<0.05)。与中高分化、TNMⅠ~Ⅱ期、无脉管癌栓的胃癌组织比较,低分化、TNMⅢ期、有脉管癌栓的胃癌组织中lncRNA ZFAS1的表达水平明显增加、miR-150-5p的表达水平降低(P<0.05)。Kaplan-Meier曲线分析显示,lncRNA ZFAS1高表达胃癌患者的累积生存率明显低于lncRNA ZFAS1低表达胃癌患者(P<0.05),不同miR-150-5p表达胃癌患者的累积生存率比较无统计学差异(P>0.05)。结论胃癌组织中lncRNA ZFAS1/miR-150-5p轴表达异常与病理特征恶化有关,lncRNA ZFAS1高表达是预后的影响因素。

Objective To study the correlation between the expression of lncRNA ZFAS1/miR-150-5p axis and the pathological characteristics,prognosis of patients undergoing radical gastrectomy.Methods From January 2015 to December 2015,the patients who underwent radical gastrectomy in General Hospital of Jizhong Energy Fengfeng Group Co.,Ltd were selected to collect the tissues of gastric cancer and adjacent tissues,the expression level of lncRNA ZFAS1 and miR-150-5p were detected and their correlation was analyzed.Overall survival was followed up,and the difference of overall survival between gastric patients with different expression of lncRNA ZFAS1,miR-150-5p was analyzed by Kaplan Meier curve.Results Compared with the adjacent tissues,the expression level of lncRNA ZFAS1 significantly increased,and the expression level of miR-150-5p significantly decreased in gastric cancer tissues(P<0.05),and the expression level of lncRNA ZFAS1 was negatively correlated with the expression level of miR-150-5p(r=-0.392,P<0.05).Compared with the moderately differentiated,well differentiated,TNMⅠ~Ⅱstage and non-vascularized tumor thrombus gastric cancer tissues,the expression level of lncRNA ZFAS1 significantly increased,and the expression level of miR-150-5p significantly decreased in poorly differentiated,TNMⅢstage and vascularized tumor thrombus gastric cancer tissues(P<0.05).Kaplan-Meier curve analysis showed that the cumulative survival rate of patients with high expression of lncRNA ZFAS1 was significantly lower than that of patients with low expression of lncRNA ZFAS1(P<0.05),there was no significant difference in the cumulative survival rate of patients with different miR-150-5p expression(P>0.05).Conclusion The abnormal expression of lncRNA ZFAS1/miR-150-5p axis in gastric cancer is related to the deterioration of pathological features,and the high expression of lncRNA ZFAS1 is a prognostic factor.
