摘要 目的揭示垂体同源配对同源结构域转录因子(PITX1)在三阴性乳腺癌(TNBC)中的表达水平及其与临床病理特征的关系,为理解这一重要转录因子在TNBC中的作用提供临床依据。方法采用免疫组织化学染色检测PITX1在120例TNBC患者病理组织中的表达情况,观察与分析PITX1表达水平与TNBC患者临床病理特征的关系,采用Kaplan-Meier法绘制患者的生存曲线并分析其与PITX1表达的相关性。结果PITX1在不同TNBC组织中的表达水平差异很大,呈现高、低表达两种不同状态,阳性信号主要位于细胞核。PITX1表达水平越低的TNBC,恶性程度越高,发生侵袭和远端转移的可能性更大。PITX1表达水平与患者总体生存率和无病生存率呈显著正相关关系。结论PITX1在TNBC病理组织中很可能发挥重要的抑癌基因作用,对其表达水平的系统评估可为临床诊治TNBC提供重要的生物学新靶点。 Objective To reveal the expression level of pituitary homeobox paired homeodomain transcription(PITX1)in triple negative breast cancer(TNBC)and its relationship with clinicopathological characteristics,so as to provide a clinical basis for understanding the role of this important transcription factor in TNBC.Methods Immunohistochemical staining was used to detect the expression of PITX1 in pathological tissues of 120 TNBC patients.The relationship between the expression level of PITX1 and the clinicopathological characteristics of TNBC patients was observed and analyzed.The survival curve of patients was drawn by Kaplan-Meier method and its correlation with the expression of PITX1 was analyzed.Results The expression levels of PITX1 in different TNBC tissues were very different,showing two different states of high and low expression,and the positive signal was mainly located in the nucleus.The lower expression level of PITX1,the higher the degree of malignancy and the greater the possibility of invasion and distal metastasis.The expression level of PITX1 was significantly positively correlated with the overall survival rate and disease-free survival rate.Conclusion PITX1 may play an important role in tumor suppressor gene in the pathological tissue of TNBC.The systematic evaluation of its expression level can provide an important new biological target for the clinical diagnosis and treatment of TNBC.
