麸炒茅苍术挥发油抗LPS诱导HCoEpiC炎症损伤的作用 被引量:8

Effect of essential oil obtained from Atractylodes lancea before and after bran-processed on lipopolysaccharide-induced inflammatory injury of human colonic epithelial cells

摘要 目的考察茅苍术挥发油抗脂多糖(LPS)诱导人结肠上皮细胞(HCoEpiC)炎症损伤的作用,并比较生品和麸炒品的作用差异。方法水蒸气蒸馏法提取茅苍术生品和麸炒品的挥发油;采用LPS造成HCoEpiC炎症损伤模型,MTT法检测细胞活力,ELISA法检测炎症相关因子IL-4,IL-6,IL-1β和TNF-α的水平;RT-PCR法检测IL-6,IL-8和TNF-αmRNA的表达;Western blot法检测IL-6和IL-8的蛋白表达量。结果与模型组比较,茅苍术生品和麸炒品挥发油均能提高HCoEpiC的细胞活力;提高抑炎因子IL-4的水平,降低促炎因子IL-6,IL-1β和TNF-α的水平;且能够降低IL-6,IL-8和TNF-αmRNA的表达和IL-6和IL-8的蛋白表达量,麸炒品的作用优于生品。结论茅苍术挥发油具有明显抗LPS诱导HCoEpiC炎症损伤的作用,麸炒后该作用增强。 Objective This study aimed at investigating the effect of Atractylodes lancea(AL)on inflammatory injury of HCoEpiC by LPS and comparing the effects between the crude AL and AL processed with bran.Methods Water vapor distillation was used to extract the exxential oil from the crude and processed AL.The cell viability was detected by an MTT assay.The levels of IL-4,IL-6,IL-1βand TNF-αwere determined by ELISA,and the mRNA expressions of IL-6,IL-8 and TNF-αwere determined by quantitative real-time PCR.Meanwhile,the expressions of IL-6 and IL-8 proteins were determined by Western blot.Results Compared with the model group,the cell viability was increased,and the production of IL-4 was significantly stimulated.Furthermore,the productions of IL-6,IL-1β,TNF-αand the mRNA expressions of IL-6,IL-8,TNF-αand the expressions of IL-6 and IL-8 proteins were notably inhibited by essential oil obtained from both the crude AL and the bran-processed AL.The essential oil from the processed AL was more effective compared with that from crude AL.Conclusion The essertial oil from AL had satisfactory effects in treatment of inflammatory injury of HCoEpiC induced by LPS,and the effects were enhanced after processed with wheat bran.
