扶正抑瘤汤对肺癌荷瘤小鼠肿瘤生长、免疫微环境的影响 被引量:3

Effect of Fuzheng Yiliu Decoction on Tumor Growth and Immune Microenvironment in Nude Mice Bearing Lung Cancer

摘要 目的研究扶正抑瘤汤对肺癌荷瘤小鼠肿瘤生长、免疫微环境的影响。方法将50只清洁级C57BL/6小鼠建立肺癌荷瘤小鼠模型,50只小鼠造模成功48只,其中模型组9只,顺铂组9只,扶正抑瘤汤高、中、低剂量组均10只。扶正抑瘤汤高、中、低剂量组小鼠分别灌胃24 g/kg、12 g/kg、6 g/kg扶正抑瘤汤;顺铂组腹腔注射4 mg/kg顺铂;模型组灌胃等体积生理盐水。比较各组小鼠肿瘤生长抑制率,各组小鼠血清TGF-β、白细胞介素2(IL-2)、干扰素(IFN-γ)浓度,比较各组小鼠肿瘤组织中及程序性死亡受体1(PD-L1)、CD4CD25+Treg细胞表达情况,Bax、Bcl-2蛋白表达水平及Bax、Bcl-2 mRNA相对表达量。结果各组小鼠肿瘤生长抑制率、IL-2、IFN-γ浓度及肿瘤组织中Bax mRNA、Bax蛋白相对表达水平均高于模型组,差异有统计学意义(P<0.05);各组小鼠血清TGF-β浓度及PD-L1表达、CD4+CD25+Treg细胞占比、Bcl-2 mRNA、Bcl-2蛋白相对表达水平均低于模型组,差异有统计学意义(P<0.05);扶正抑瘤汤的作用呈剂量依赖性,且扶正抑瘤汤高剂量组优于顺铂组。结论扶正抑瘤汤对肺癌荷瘤小鼠肿瘤生长具有抑制作用,可改善肿瘤免疫微环境。 Objective To study the effect of Fuzheng Yiliu Decoction on tumor growth and immune microenvironment in mice bearing lung cancer.Methods 50 clean level C57BL/6 mice were used to establish lung cancer tumor-bearing mouse models.48 mice were successfully modeled,which were randomly divided into 5 groups:9 in the model group and 9 in the cisplatin group,10 in the Fuzheng Yiliu Decoction high,medium and low dose,respectively.Fuzheng Yiliu Decoction high,medium,and low dose groups of mice were given 24 g/kg,12 g/kg,and 6 g/kg Fuzheng Yiliu Decoction by gavaged,and the cisplatin group was intraperitoneally injected with 4 mg/kg cisplatin.The model group was given an equal volume of normal saline by gavaged.Results The tumor growth inhibition rates,interleukin 2(IL-2),interferonγ(IFN-γ)and relative expression levels of Bax mRNA and protein in tumor tissues in each group were higher than the model group(P<0.05),while transforming growth factorβ(TGF-β)levels,programmed death receptor 1(PD-L1),CD4+,CD25+Treg proportions and relative expression levels of Bcl-2 mRNA and protein in the each groups of mice were lower than the model group(P<0.05).The effect of Fuzheng Yiliu Decoction was dose-dependent,and the Fuzheng Yiliu Decoction high dose group was better than cisplatin group.Conclusion Fuzheng Yiliu Decoction can inhibit tumor growth and improve tumor immune microenvironment in lung cancer bearing mice.
