摘要
目的探讨FOXK1在食管癌组织中的表达及与食管癌细胞增生和侵袭转移的关系。方法选取2018年3月至2019年9月间洛阳市中心医院收治的95例食管癌患者组织样本,采用蛋白免疫印迹法检测食管癌细胞系及组织中FOXK1表达,通过免疫组化对食管癌组织芯片进行分析。构建FOXK1稳定过表达株,通过细胞增殖实验、划痕实验及transwell侵袭小室实验探究FOXK1表达与食管癌细胞恶行生物学行为的关系。通过免疫印迹检测上皮间质转化(EMT)相关蛋白表达,探索FOXK1与食管癌细胞EMT的潜在关系。结果FOXK1在食管癌细胞系及食管癌组织中表达上调,组织芯片分析也证实FOXK1在食管癌组织中为高表达,且与患者的生存预后相关。过表达FOXK1后,食管癌细胞增殖能力、划痕愈合能力和侵袭能力增强,且FOXK1参与TGF-β1诱导的EMT。结论FOXK1在食管癌的增殖、侵袭及转移上发挥重要作用,为食管癌的早期诊断、预后判断和治疗提供新思路。
Objective To investigate the expression of FOXK1 in esophageal cancer and its relationship with the proliferation,invasion and metastasis of cells.Methods Tissue samples were collected from 95 patients with esophageal cancer at Luoyang Central Hospital between March 2018 and September 2019.The expression of FOXK1 in esophageal cancer cells and tissues were detected by western blot,and immunohistochemistry was used to analyze the tissue microarray of esophageal cancer.The stable FOXK1-overexpression strains were established and cell proliferation bioassay,scratch test and Transwell invasion test were used to explore the relationship between overexpression of FOXK1 and malignant biological behaviors of esophageal cancer cells.Western blot was used to detect the expression of epithelial-mesenchymal-transition(EMT)-related protein to explore the potential relationship between FOXK1 and EMT in esophageal cancer cells.Results The expression of FOXK1 was up-regulated in esophageal cancer cells and tissues and the tissue microarray analysis also confirmed that FOXK1 was highly expressed in esophageal cancer tissues and was significantly associated with the prognosis of patients.The ability of proliferation,invasion and metastasis were enhanced in FOXK1-overexpression cancer cells and FOXK1 participated in TGF-β1-induced EMT.Conclusion FOXK1 may play an important role in the proliferation,invasion and metastasis of esophageal cancer providing new ideas for early diagnosis,prognosis and treatment of esophageal cancer.
