摘要
目的:探讨姜黄素对耐5氟尿嘧啶和奥沙利铂的结肠癌细胞的抑制作用。方法:大剂量冲击方法联合剂量梯度法建立结肠癌耐5氟尿嘧啶和奥沙利铂的细胞,Real-time PCR方法检测耐药细胞的干性基因表达情况,MTT方法检测姜黄素对耐药细胞的增殖抑制作用。结果:成功建立结肠癌耐5氟尿嘧啶和奥沙利铂细胞,其干细胞相关基因如CD133、CD24、ALDH1、OCT-4、SOX-2和Nanog1表达上调,提示其干性增高可能是结肠癌耐药的一个主要因素,姜黄素对两种耐药结肠癌细胞的增殖具有明显抑制作用,且能抑制耐药细胞干细胞标志物的表达。结论:姜黄素可有效抑制耐药结肠癌细胞,有望成为结肠癌治疗的辅助性药物。
Objective To investigate the inhibitory effect of curcumin on colon cancer cells resistant to 5-fluorouracil and oxaliplatin.Methods 5-fluorouracil and oxaliplatin resistant colon cancer cells lines were established by high-dose shock combined with dose gradient method.The expression of stemness genes was detected by real-time PCR,and the proliferation inhibition of curcumin on drug-resistant cell lines was detected by MTT method.Results 5-fluorouracil-resistant and oxaliplatin resistant colon cancer cell lines were successfully established,and the stem cell related genes were upregulated such as CD133,CD24,ALDH1,OCT-4,SOX-2 and Nanog1,suggesting that the increased stemness may be a major factor of drug resistance.Curcumin can effectively inhibit the proliferation of drug-resistant cell lines and inhibit the expression of stem cell markers of drug-resistant cells.Conclusion Curcumin can effectively inhibit drug-resistant colon cancer cells,and is expected to become an adjuvant drug in the treatment of colon cancer.
