葫芦素B通过阻断STAT3活化抑制胃癌发生发展的作用机制研究

Cucurbitacin B Inhibits Gastric Cancer Progression by Suppressing STAT3 Activation

目的:探讨葫芦素B(Cucurbitacin B,CuB)抑制胃癌细胞发生发展的分子机制。方法:MTT法、克隆形成实验检测CuB对胃癌细胞MGC803和SGC7901增殖的影响;Transwell细胞侵袭实验检测细胞侵袭能力;Annexin V-FITC/PI双染法检测细胞凋亡;SwissTargetPrediction在线预测与CuB相结合的关键因子;Western blot法检测信号通路蛋白表达情况;分子对接模拟CuB与STAT3的结合作用。结果:CuB对胃癌细胞的增殖具有显著的抑制作用并诱导胃癌细胞凋亡。在线预测发现STAT3是CuB潜在结合靶点,分子对接发现CuB与STAT3直接结合。结论:CuB通过靶向STAT3抑制胃癌细胞增殖并诱导凋亡。

Objective To investigate the molecular mechanism of cucurbitacin B(CuB)inhibiting the occurrence and development of gastric cancer cells.Methods MTT assay and clone forming assay were used to detect proliferation of gastric cancer cells MGC803 and SGC7901.Transwell assay was used to detect cell invasion ability of GC cells.AnnexinV-FITC/PI double staining method was used to detect apoptosis.SwissTargetPrediction online prediction was used to detect the key factors combined with Cub.Western blot was used to detect the expression of STAT3 signaling pathway proteins.Molecular docking was used to simulate the combination of CuB and STAT3.Results CuB had a significant inhibitory effect on the proliferation and induced apoptosis of gastric cancer cells.The online prediction found that STAT3 was a potential target of CuB,and molecular docking found that Cub directly bound to STAT3.Conclusion CuB can inhibite proliferation and induce apoptosis of gastric cancer cells by directly targeting STAT3.
