摘要
【目的】建立一套党武辣椒最优快繁技术体系,为实现贵安地区种苗快速繁殖和大面积种植提供技术参考。【方法】以党武辣椒为研究对象,经25℃无菌条件发芽生长,选取无菌苗去顶芽茎尖为生长点,以MS为基本培养基,通过添加不同浓度的6-BA、NAA、GA 3、IAA等生长调节剂进行筛选试验,取15~30 d苗龄的无菌苗进行梯度试验。【结果】MS+0.05 mg/L NAA+0.5 mg/L 6-BA是直接成苗途径侧芽分化的最佳配方;MS+0.05~0.07 mg/L NAA+5.0 mg/L 6-BA适宜于愈伤组织途径不定芽的侧芽分化;适合党武辣椒最佳侧芽分化苗龄为25~30 d;MS+0.01~0.05 mg/L NAA适宜根的诱导生长;生根后进行驯化移栽的成活率高于95%。【结论】党武辣椒快繁种苗可通过直接成苗途径与愈伤途径获得,种苗分化率高、种质好、成活率高。
【Objective】A set of optimal rapid propagation technological system for seedling production of Dangwu Pepper is established to provide a reference for rapid propagation and large-scale planting of pepper seedlings in Gui’an,Guizhou.【Method】The stem tip segments of aseptic seedlings germinated from DangwuPepper seeds at 25℃are used as explants and then the explants are cultured on MS media with different plant growth regulators combination of 6-BA,NAA,GA 3,IAA,2,4-D and AgNO 3 to screen the optimal media for lateral bud differentiation of the explants.The explants from sterile seedlings with different seedling age(15 d,20 d,25 d and 30 d)are cultured on three screened optimal media respectively to discuss the relationship between optimal lateral bud differentiation rate and seedling age.【Result】MS+NAA 0.05 mg/L+6-BA 0.5 mg/L is the optimal medium for lateral bud differentiation of the explants throughthe direct seedling path.MS+NAA 0.05~0.07 mg/L+6-BA 5.0 mg/L is the optimal medium for lateral bud differentiation of adventitious buds through the callus tissue path.The optimal sterile seedling age for lateral bud differentiation is 25~30 d.MS+NAA 0.01~0.05 mg/L issuitable for root inductionand growth of the explants and the acclimatization and transplanting survival rate of the explants with roots can reach above 95%.【Conclusion】The seedlings with higher differentiation rate,good germplasm and higher survival rate can be obtained through direct seedling and callus tissue paths in rapid propagation of Dangwu Pepper.
