摘要
目的探讨微小RNA-1265(miR-1265)在鼻咽癌中的表达及诊断价值。方法选取2018年1月至2019年8月于该院治疗的鼻咽癌患者60例作为研究组,鼻咽部良性肿瘤患者60例作为对照组,收集两组患者的病理组织和血清。采用PCR法检测两组患者病理组织和血清中的miR-1265相对表达水平,比较不同临床病理参数鼻咽癌患者miR-1265的相对表达水平。采用受试者工作曲线(ROC曲线)分析miR-1265对鼻咽癌的诊断价值,采用TargetScan预测靶基因并用双荧光素酶报告基因分析实验进行验证。采用Pearson相关分析miR-1265和MAT1表达的相关性。结果研究组病理组织中miR-1265的相对表达水平高于对照组(P<0.05)。研究组患者血清中miR-1265的相对表达水平高于对照组(P<0.05)。不同性别、年龄及病理类型患者间miR-1265的表达水平差异无统计学意义(P>0.05),不同TNM分期、病理分期和淋巴结转移的患者miR-1265表达水平差异有统计学意义(P<0.05)。当病理组织miR-1265截断值为0.895时,其诊断鼻咽癌的曲线下面积(AUC)为0.917(95%CI:0.878~0.946),灵敏度为87.3%,特异度为91.3%。当血清miR-1265截断值为0.515时,其诊断鼻咽癌的AUC为0.717(95%CI:0.678~0.806),灵敏度为67.3%,特异度为71.3%。双荧光素酶报告基因分析实验证实miR-1265的靶基因为MAT1。鼻咽癌组织中MAT1 mRNA与miR-1265的相对表达水平呈显著负相关(r=-0.943,P<0.001)。结论鼻咽癌组织和血清中miR-1265表达上调,与鼻咽癌进展密切相关,具有区分鼻咽癌和鼻咽部良性肿瘤的价值。miR-1265靶基因是MAT1基因,可能成为鼻咽癌潜在治疗靶点。
Objective To investigate the expression and diagnostic value of miR-1265 in nasopharyngeal carcinoma(NPC).Methods From January 2018 to August 2019,60 patients with NPC and 60 patients with benign tumor of nasopharynx were enrolled in the study as study group and control group,respectively.The pathological tissue and serum samples of the two groups were collected.The relative expression levels of miR-1265 in pathological tissue and serum of the two groups was detected by PCR.The difference of relative expression levels of miR-1265 in patients with NPC with different clinical pathological parameters was compared.Receiver operating characteristic curve(ROC curve)was used to analyze the value of miR-1265 in the diagnosis of NPC.The target gene of miR-1265 was predicted by using TargetScan and verified by double luciferase experiment.Pearson correlation was used to analyze the correlation between miR-1265 and MAT1 expression.Results The relative expression of miR-1265 in pathological tissues of the study group was higher than that of the control group(P<0.05).The relative expression of miR-1265 in the serum of the study group was higher than that of the control group(P<0.05).There was no significant difference in the expression of miR-1265 in patients with different gender,age and pathological type(P>0.05),but there was significant difference in the expression of miR-1265 in patients with different TNM stage,pathological stage and condition of lymph node metastasis(P<0.05).When the cut-off value of miR-1265 was 0.895,the AUC for NPC diagnosis was 0.917(95%CI:0.878-0.946),sensitivity was 87.3%,specificity was 91.3%.When the cut-off value of miR-1265 was 0.515,the AUC of NPC was 0.717(95%CI:0.678-0.806),sensitivity was 67.3%,specificity was 71.3%.The target of miR-1265 is MAT1.There was a significant negative correlation between the relative expression of MAT1 mRNA and miR-1265 in NPC(r=-0.943,P<0.001).Conclusion The expression of miR-1265 is up-regulated in NPC tissues and serum.It has the diagnostic value of distinguishing NPC from benign tumors in nasopharynx.The target gene of miR-1265 is MAT1 gene,which may become a therapeutic target for NPC.
