摘要
目的探究体外冲击波联合玻璃酸钠对兔膝骨关节炎软骨组织的治疗作用及可能机制。方法 30只兔随机分成对照组、模型组及治疗组,每组10只。HE染色观察软骨组织形态学变化;免疫荧光检测软骨组织Runx2和Osteocalcin蛋白表达;TUNEL方法检测软骨组织细胞凋亡;Western blot检测β-catenin、Wnt4a、MMP-1和MMP-13蛋白表达。结果与对照组相比,模型组软骨组织形态学病理变化明显,细胞凋亡水平升高,Runx2、Osteocalcin、β-catenin、Wnt4a、MMP-1和MMP-13蛋白表达水平升高。体外冲击波联合玻璃酸钠改善软骨组织形态,降低软骨组织细胞凋亡、Runx2、Osteocalcin、β-catenin、Wnt4a、MMP-1和MMP-13蛋白表达水平。结论体外冲击波联合玻璃酸钠促进兔膝骨关节炎软骨修复,其作用机制可能与Wnt/β-catenin通路有关。
Objective To explore the therapeutic effect of extracorporeal shock wave combined with sodium hyaluronate on cartilage tissue of rabbit knee osteoarthritis and its possible mechanism. Methods Thirty rabbits were randomly divided into control group, model group and treatment group, with 10 rabbits in each group. The histological changes of cartilage were observed by HE staining. The expressions of Runx2 and Osteocalcin in cartilage were detected by immunofluorescence. TUNEL assay was used to detect the apoptosis of cartilage cells. The protein expressions of β-catenin, Wnt4 a, MMP-1 and MMP-13 were detected by Western Blot. Results Compared with the control group, the histological and pathological changes of cartilage in the model group were obvious, the level of apoptosis was increased, and the protein expression levels of Runx2, Osteocalcin, β-catenin, Wnt4 a, MMP-1 and MMP-13 were increased. Extracorporeal shock wave combined with sodium hyaluronate improved the morphology of cartilage tissue, and decreased the levels of cartilage apoptosis, Runx2, Osteocalcin, β-catenin, Wnt4 a, MMP-1 and MMP-13 protein expressions. Conclusion Extracorporeal shock wave combined with sodium hyaluronate can promote cartilage repair in rabbit knee osteoarthritis, and the mechanism of action may be related to Wnt/β-catenin pathway.
