摘要
目的研究LncRNA AL022344.7在乳腺癌临床标本数据库和细胞中的差异表达及其对乳腺癌细胞生物学表型的调控作用。方法下载TCGA数据库,采用R语言分析发现LncRNA AL022344.7在乳腺癌组织中差异表达。qRT-PCR法检测多株乳腺癌细胞中LncRNA AL022344.7的表达情况,在MCF-10细胞中敲减LncRNA AL022344.7,CCK-8实验检测细胞增殖情况,Transwell小室实验和划痕愈合实验检测细胞迁移情况,Western blot检测细胞上皮-间质转化相关蛋白的表达变化。结果 LncRNA AL022344.7在多株乳腺癌细胞中均表达下调。LncRNA AL022344.7在MCF-10细胞中敲减后,细胞的增殖能力显著下降,侵袭和转移能力也下调。进一步研究发现,敲减LncRNA AL022344.7可以抑制E-钙黏蛋白(E-cadherin)表达,上调波形蛋白(Vimentin)的表达。结论 LncRNA AL022344.7在乳腺癌组织和细胞中表达上调,LncRNA AL022344.7敲减可以抑制乳腺癌细胞增殖、侵袭、转移和EMT。
Objective To study the differential expression of LncRNA AL022344.7 in breast cancer clinical specimen databases and cells and its regulatory effect on the biological phenotype of breast cancer cells. Methods Through downloading the TCGA database, using R language analysis, LncRNA AL022344.7 was demonstrated to be differentially expressed in breast cancer tissues. qRT-PCR method was used to detect the expression of LncRNA AL022344.7 in multiple breast cancer cells. After LncRNA AL022344.7 was knocked down in MCF-10 cells, CCK-8 experiment was performed to detect cell proliferation, Transwell assay and scratch healing experiment were used to detect cell migration;Western blot was performed to detect cell epithelial-mesenchymal transition related protein expression. Results The expression of LncRNA AL022344.7 was down-regulated in multiple breast cancer cells. At the same time, it was found that after knocking down LncRNA AL022344.7 in MCF-10 cells, the proliferation ability of the cells decreased significantly, and the invasion and metastasis ability of the cells was also down-regulated. Further research found that knockdown of LncRNA AL022344.7 can inhibit the expression of E-cadherin and up-regulate the expression of vimentin. Conclusion The expression of LncRNA AL022344.7 is up-regulated in breast cancer tissues and cells. Knockdown of LncRNA AL022344.7 can inhibit breast cancer cell proliferation, invasion and metastasis and EMT.
