活化肝星状细胞促进肠癌奥沙利铂耐药 被引量:2

Activated hepatic stellate cells on oxaliplatin resistance in colorectal cancer

摘要 目的研究活化肝星状细胞促进肠癌奥沙利铂耐药。方法 Western blot方法及免疫荧光方法检测肝星状细胞α-SMA及CollagenI蛋白表达,同时Western blot方法检测肠癌细胞上皮间质转化(Epithelialmesenchymal transition, EMT)相关蛋白Vimentin与E-cadherin的表达,应用集落形成实验检测肠癌细胞增殖能力,Transwell实验检测肠癌细胞迁移能力,MTT实验验证肠癌细胞的奥沙利铂敏感性。结果肝星状细胞α-SMA及CollagenI高表达;活化肝星状细胞促进Caco2及RKO肠癌细胞增殖能力、迁移能力及奥沙利铂耐药。Caco2及RKO肠癌细胞镜下细胞形态发生改变,EMT相关蛋白Vimentin表达增加,E-cadherin表达降低。结论活化肝星状细胞促使肠癌细胞发生EMT从而对奥沙利铂耐药。 Objective To study activated hepatic stellate cells(aHSC)on oxaliplatin resistance in colorectal cancer. Methods The content of α-SMA and collagenI protein in HSCs were detected by Western blot and immunofluorescence. The expression of EMT associated protein(Vimentin,E-cadherin) in colorectal cancer cellswere detected by Western blot. The proliferation ability of colorectal cancer cells was detected by colony formation test.The migration ability of colorectal cancer cells was detected by transwell test. The sensitivity of oxaliplatin was verified by MTT test. Results α-SMA and CollagenI were highly expressed in aHSC. aHSC promoted the proliferation, migration and oxaliplatin resistance in Caco2 and RKO cells. The morphology of Caco2 and RKO cells changed under microscope. The expression of EMT-related protein vimentin increased and the expression of E-cadherin decreased. Conclusion aHSC promoted the development of EMT in colorectal cancer cells which resulted in oxaliplatin resistance.
