高表达miR-20b对甲状腺癌TPC-1细胞增殖和侵袭的影响及可能机制

The effect high expression of miR-20b on proliferation and invasion of TPC-1 cells in thyroid cancer and its possible mechanism

摘要 目的探究高表达miR-20b对甲状腺癌TPC-1细胞增殖和侵袭的影响及其可能机制。方法将TPC-1细胞分为空白对照组、 miR-NC组(转染阴性对照miR-NC)和miR-20b模拟物组(转染miR-20b mimics),实时定量PCR和Western blot检测各组细胞miR-20b和CCND1的表达,双荧光素酶报告基因实验分析miR-20b和CCND1的靶向关系。MTT方法检测各组TPC-1细胞第24h、48h、72h的增殖能力,Transwell实验检测各组TPC-1细胞的侵袭能力。结果实时定量PCR和Western blot结果显示,转染miR-20b模拟物后,TPC-1细胞中miR-20b的表达升高,CCND1的表达下降(P<0.05),生物信息学和双荧光素酶实验表明CCND1是miR-20b的靶基因。转染miR-20b模拟物后,TPC-1细胞的增殖和侵袭能力下降(P<0.05)。结论高表达miR-20b可抑制甲状腺癌细胞TPC-1的增殖和侵袭能力,其机制可能与CCND1表达受到抑制有关。 Objective To investigate the effect and mechanism of miR-20 b on the proliferation and invasion of TPC-1 cells. Methods The TPC-1 cells were divided into blank control group, miR-NC group(negative control miR-NC) and miR-20 b mimics group(miR-20 b mimics). The expression of miR-20 b and CCND1 was detected by quantitative realtime PCR and Western blot, and the target relationship between miR-20 b and CCND1 was verified by double luciferase assay. The proliferation ability of TPC-1 cells was detected by MTT, and the invasion ability of TPC-1 cells was detected by Transwell assay. Results The results of real-time quantitative PCR and Western blot showed that the expression of miR-20 b in TPC-1 cells was increased and CCND1 was decreased(P<0.05). After transfection of miR-20 b mimics, the proliferation and invasion ability of TPC-1 cells decreased(P<0.05). Conclusion high expression of miR-20 b inhibited the proliferation and invasion of TPC-1 cells, which might be related to down-regulation of CCND1 expression.
