木棉花水提物对人肺癌NCI-H460裸鼠移植瘤抑制作用及机制研究 被引量:3

摘要 目的研究木棉花水提物对人肺癌NCI-H460细胞接种的裸鼠移植瘤的抑制作用及机制。方法对人肺癌NCI-H460细胞接种成功的裸鼠灌胃给予木棉花水提物(1.2 g·kg^(-1)、0.6 g·kg^(-1),以生药量计),观察裸鼠的一般状态并检测各组移植瘤体积变化;给药14 d后处死裸鼠,剥离瘤体称重并计算抑制率;酶联免疫吸附法(enzyme linked immuno sorbent assay, ELISA)测定血清中白介素6(interleukin 6,IL-6)和肿瘤坏死因子α(tumor necrosis factorα,TNF-α)的水平。比色法测定瘤组织中丙二醛(malondialdehyde, MDA)和超氧化物歧化酶(super oxide dismutase, SOD)的含量;苏木精-伊红(hematoxylin-eosin, HE)染色法观察瘤组织细胞形态学变化;免疫组化法检测瘤组织中p53、Bax、Bcl-2、Caspase-9、Caspase-3蛋白表达水平。结果与模型组比较,木棉花水提物高剂量组瘤体积明显减小,瘤质量明显减轻,抑瘤率达59.7%;血清中IL-6、TNF-α显著升高,瘤组织中SOD活力明显升高,MDA含量显著降低;上调p53、Bax、Caspase-9、Caspase-3蛋白表达水平,下调Bcl-2蛋白表达水平。结论木棉花水提物对人肺癌NCI-H460细胞接种的裸鼠移植瘤生长具有明显的抑制作用。其作用机制可能是改善机体的免疫调节能力,提高抗氧化能力和诱导细胞凋亡。 Objective To study the inhibitory effect and mechanism of aqueous extract of Bombax malabaricum flowers on human lung cancer NCI-H460 tumor transplanted in nude mice.Methods Aqueous extract of Bombax malabaricum flowers(1.2 g·kg^(-1),0.6 g·kg^(-1)) was given to the nude mice with human lung cancer NCI-H460 cells by gavage, to observe the general state of nude mice and detect the volume change of transplanted tumor in each group.The nude mice were sacrificed after administration for 14 days, the tumors were removed and weighed, and the inhibition rate was calculated.The levels of interleukin 6(IL-6) and tumor necrosis factor α(TNF-α) in serum were determined by enzyme linked immuno sorbent assay(ELISA).The contents of malondialdehyde(MDA) and super oxide dismutase(SOD) were measured by colorimetry, the morphological changes of tumor tissues were observed by hematoxylin-eosin(HE) staining, and the expression levels of p53,Bax, Bcl-2,caspase-9 and caspase-3 were detected by immunohistochemistry.Results Compared with the model group, the tumor volume was significantly reduced and tumor mass was significantly reduced in the aqueous extract with high-dose group, and the tumor inhibition rate was 59.7%.The levels of IL-6 and TNF-α in serum were significantly increased, and the activity of SOD in tumor tissue was significantly increased, while the content of MDA was significantly decreased.The expression levels of p53,Bax, caspase-9,caspase-3 protein were up-regulated, and the expression level of Bcl-2 protein was down-regulated.Conclusion The aqueous extract of Bombax malabaricum flowers can inhibit the growth of human lung cancer NCI-H460 tumor transplanted in nude mice, and the action mechanism may be to improve the immune regulation ability of organism, enhance the antioxidant capacity and induce apoptosis.
