摘要
目的建立刺柏叶的高效液相色谱(HPLC)指纹图谱,鉴别其易混淆品种。方法采用HPLC方法测定12批刺柏叶药材和4批易混淆品种,对测得的色谱数据进行相似度评价,并对12批刺柏叶进行聚类分析和主成分分析。结果建立的刺柏叶指纹图谱标定了18个共有峰,其中指认13号峰为穗花杉双黄酮色谱峰;12批刺柏叶与对照指纹图谱的相似度范围为0.962~0.996,4批易混淆品种与对照指纹图谱的相似度范围为0.580~0.951;聚类分析中12批刺柏叶分为2类;主成分分析中提取出累计方差贡献率为90.777%的5个主成分,以这5个主成分计算所有批次刺柏叶的综合得分,得到刺柏叶药材的质量排序结果。结论建立的HPLC指纹图谱可以有效的鉴别刺柏叶正品和易混淆品种,结合聚类分析和主成分分析可以对刺柏叶药材的质量进行客观、有效的评价。
Objective To establish High Performance Liquid Chromatography(HPLC) fingerprint of Juniperus rigida leaves and identify its confused varieties. Methods 12 batches of Juniperus rigida leaves and 4 batches of confused varieties are determined by HPLC, and the similarity of chromatographic data is evaluated. Cluster analysis and principal component analysis are performed on 12 batches of Juniperus rigida leaves. Results 18 common peaks are identified in the fingerprint of Juniperus rigida leaves, among which peak 13 is amentoflavone;the similarity ranges between 12 batches of Juniperus rigida leaves and the reference fingerprint are 0.962 to 0.996, and the similarity ranges between 4 batches of confused varieties and the reference fingerprint are 0.580 to 0.951;cluster analysis indicates that 12 batches of Juniperus rigida leaves are divided into 2 groups;principal component analysis shows that 5 principal components with cumulative variance contribution rate of 90.777% are extracted to calculate the comprehensive scores of all batches of Juniperus rigida leaves, of which the quality is ranked. Conclusion The established HPLC fingerprint can effectively identify the genuine and confused varieties of Juniperus rigida leaves, and the quality of Juniperus rigida leaves can be objectively and effectively evaluated combined with cluster analysis and principal component analysis.
