摘要
目的了解腓骨蛋白-1(FBLN-1)在2型糖尿病患者牙槽骨骨髓间充质干细胞(BMSCs)中的表达及其对骨髓间充质干细胞的生物学行为影响。方法原代培养口腔种植患者的牙槽骨BMSCs,分为3组:种植成功的糖尿病患者(DM-S组)、种植失败的糖尿病患者(DM-F组)以及非糖尿病种植成功患者(Nor组)。MTT法比较3组细胞增殖能力的差异;划痕实验观察细胞的迁移能力;碱性磷酸酶实验测定3组细胞的细胞活性差异;Western-blot检测FBLN-1在3组BMSCs中的表达差异。结果BMSCs增殖能力,Nor组明显优于DM-S组和DM-F组(P<0.05),但DM-S组和DM-F组无统计学差异;6 h细胞迁移能力测定,发现Nor组BMSCs迁移距离明显大于DM-S组(P<0.05),DM-S组BMSCS迁移距离明显大于DM-F组BMSCs(P<0.05);在各时间点检测的ALP活性,Nor组BMSCs最高(P<0.05),DM-S组BMSCs次之(P<0.05),DM-F组BMSCs最低(P<0.05);FBLN-1表达,与Nor组相比DM-S组和DM-F两组均显著下调(P<0.05),且与DM-S组相比,DM-F组进一步下调(P<0.05)。结论与非糖尿病患者相比,糖尿病患者牙槽突BMSCs中FBLN-1的表达显著下调。糖尿病种植失败患者的FBLN-1明显低于糖尿病成功患者,表明FBLN-1在种植体愈合过程中可能起重要作用。
Objective To investigate the expression of FBLN-1 in alveolar bone marrow mesenchymal stem cells of type 2 diabetic patients.Methods Bone marrow stem cells(BMSCs)were harvested from patients receiving dental implantation.The three groups were successfully implanted diabetic patients(group one),failed cases(group two),and successfully implanted non-diabetic patients(group three).The growth curve of the three groups of cells was measured by MTT method,the migration ability of the cells was observed by the scratch test.The expression of FBLN-1 in the three groups of BMSCs was detected by Western-blot,and the activity of alkaline phosphatase(ALP)was detected in the three groups.Results There were significant differences in cell proliferation ability,migration ability and ALP activity among the three groups(P<0.05).In addition,the expression level of FBLN-1 in group two was significantly lower than that in group one.Conclusions The expression of FBLN-1 in the alveolar bone marrow mesenchymal stem cells of type 2 diabetic patients with failed implantation decreased,indicating that fbln-1 may play an important role in the process of implant healing.
