富硒中药治疗慢性非细菌性前列腺炎大鼠的实验研究 被引量:3

摘要 目的探讨富硒中药对慢性非细菌性前列腺炎大鼠的治疗效果及其作用机制。方法选取2019年6月购入的40只8~10周龄的SPF级雄性SD大鼠作为研究对象。随机分成8笼,留1笼作为空白对照组,其余各笼大鼠采用去势(双侧睾丸切除)联合皮下注射雌二醇,诱导建立动物模型。建模成功后将大鼠按治疗方法分别设定为:模型组、中药组、低硒组、高硒组,各组均以灌胃给药的方式治疗,每次的灌胃剂量为1mL/100g·bw,连续灌胃14d,末次给药12h后称重处死,取前列腺标本,采用酶联免疫吸附试验法检测肿瘤坏死因子-α(TNF-α)、白细胞介素-1(IL-1)、诱导型一氧化氮合酶(iNOS)的含量,制备病理切片,观察腺体病理改变及炎症情况。结果经连续14d灌胃治疗后,模型组大鼠前列腺组织检测结果中TNF-α、IL-1、iNOS的含量明显高于其他组,差异具有统计学意义(P<0.05);同时,显微镜下病理切片提示前列腺腺体结构不完整,腺上皮萎缩,管腔狭窄,腺体及周围肌层有明显炎症细胞浸润。与模型组比较,普通中药组TNF-α、IL-1、iNOS的含量降低,提示表达减弱,差异具有统计学意义(P<0.05),病理切片结果显示炎症减轻。与普通中药组比较,富硒中药治疗组检测结果中TNF-α、IL-1、iNOS的含量降低,差异具有统计学意义(P<0.05),其中高硒组降低程度更大,病理切片显示炎症改善情况更好。结论相比普通中药,富硒中药更能有效减轻慢性非细菌性前列腺炎的炎症反应,降低TNF-α、IL-1、iNOS的含量及表达水平,且高硒中药治疗慢性非细菌性前列腺炎疗效更好。 Objective To investigate the therapeutic effect of selenium enriched traditional Chinese medicine on chronic nonbacterial prostatitis in rats. Methods 40 SPF male SD rats aged 8 to 10 weeks purchased in June 2019 were randomly divided into 8 cages, and 1 cage was left as the blank control group. The other groups were castrated(bilateral testis resection) combined with subcutaneous injection of estradiol to establish the animal model. After the model was successfully prepared, the rats were divided into four groups according to the treatment methods: the model group, the Chinese medicine group, the low selenium group and the high selenium group. All were treated by intragastric administration(1 mL/100 g·bw for each time) for 14 days, and were weighed and killed 12 hours after the last administration. Prostate samples were taken. The tumor necrosis factor-α(TNF-α), interleukin-1(IL-1) and inducible nitric oxide synthase(iNOS) were detected by ELISA. Pathological sections were prepared to observe the pathological changes and inflammation of the glands. Results After 14 days of gavage treatment, the contents of TNF-α, IL-1 and iNOS in prostate tissue of SD rats in model group were significantly higher than those in other groups, the difference was statistically significant(P<0.05);at the same time, under the pathological section microscope, the gland lumen was convex and narrow, without obvious secretion, with obvious inflammatory cell infiltration. The contents of TNF-α, IL-1, iNOS in prostate tissue of the Chinese medicine group was lower than that of the model group, the difference was statistically significant(P<0.05), and the pathological section of the group showed that the inflammation was reduced. Compared with the traditional Chinese medicine group, the contents of TNF-α, IL-1, iNOS in the selenium enriched traditional Chinese medicine group decreased, the difference was statistically significant(P<0.05), among them, the high selenium group decreased more significantly than the low selenium group, and the pathological section showed that the inflammation improved better. Conclusions Compared with the treatment effect of selenium-free traditional Chinese medicine, selenium enriched traditional Chinese medicine can improve the inflammatory response of chronic nonbacterial prostatitis, reduce the content and expression levels of TNF-α, IL-1, iNOS, and the therapeutic effect of selenium enriched traditional Chinese medicine has better therapeutic effect on chronic nonbacterial prostatitis.
