Study on the relationship between Fractalkine and Parkinson′s disease

目的探讨帕金森病患者血清炎症物质Fractalkine的变化及其与疾病严重程度、预后的相关性。方法选取2020年5—7月就诊于该院门诊和住院的原发性帕金森病患者40例作为研究对象。另外选取40例年龄相近、性别匹配的健康受试者作为健康对照组。评估帕金森组并记录统一帕金森病分级量表(UPDRS)、改良的Hoehn-Yahr(H-Y)分级量表、年龄、性别、病程等。根据H-Y分级量表分为早期帕金森组(≤2.5级,n=25)和中晚期帕金森组(>2.5级,n=15)。采集血样10个月后,再次随访评估帕金森病患者的UPDRS评分和H-Y分级,将UPDRS总分或H-Y分级结果升高的分为预后不佳组(n=13),UPDRS总分和H-Y分级结果降低或不变的分为预后良好组(n=26)。使用双抗体夹心酶联免疫吸附法对所有受试者血清Fractalkine表达水平进行检测并比较,分析帕金森病患者血清Fractalkine表达水平与UPDRS评分、预后的相关性。结果早期帕金森组血清Fractalkine的表达水平明显低于健康对照组和中晚期帕金森组,差异均有统计学意义(P<0.05);中晚期帕金森组血清Fractalkine的表达水平和健康对照组比较,差异无统计学意义(P>0.05)。血清Fractalkine表达与UPDRS评分呈正相关(r=0.331,P<0.05),但与H-Y分级无显著线性相关(r=0.182,P>0.05)。帕金森病患者预后良好组血清Fractalkine表达水平[(2.63±4.72)ng/mL]与预后不佳组[(3.93±9.38)ng/mL]比较,差异无统计学意义(P>0.05)。结论早期帕金森病患者的血清Fractalkine表达水平明显降低,且血清Fractalkine表达可能与帕金森病患者的病情严重程度密切相关。帕金森病患者的血清Fractalkine表达水平对预后评估无明显意义。

Objective To investigate the changes of serum inflammatory substance Fractalkine in patients with Parkinson′s disease and it′s correlation with disease severity and prognosis.Methods Forty patients with primary Parkinson′s disease who admitted to the hospital from May to July 2020 were selected as the research subjects.Another 40 healthy subjects with similar age and gender matching were selected as healthy control group.The Unified Parkinson′s Disease Rating Scale(UPDRS),Hoehn-Yahr(H-Y)staging scale,age,gender and course of disease were assessed and recorded for the Parkinson′s group.According to H-Y staging scale,the patients were divided into early-stage Parkinson′s group(≤2.5 grade,n=25)and middle-and advanced-stage Parkinson′s group(>2.5 grade,n=15).UPDRS score and H-Y staging were assessed again at 10 months after blood sampling.Patients with increased UPDRS score or H-Y staging were divided into poor prognosis group(n=13),and those with reduced or unchanged UPDRS score and H-Y staging were divided into good prognosis group(n=26).The serum Fractalkine level of all subjects was measured and compared by double-antibody ELISA,and the correlation between the serum Fractalkine level and UPDRS score and prognosis of patients with Parkinson′s disease was analyzed.Results The expression level of serum Fractalkine in early-stage Parkinson′s group was significantly lower than that in healthy control group and middle-and advanced-stage group,the differences were statistically significant(P<0.05);the expression level of serum Fractalkine in middle-and advanced-stage group and health control group had no statistically significant difference(P>0.05).The expression level of serum Fractalkine was positively correlated with UPDRS score(r=0.331,P<0.05),but there was no significant linear correlation with H-Y staging(r=0.182,P>0.05).There was no significant difference in the expression level of serum Fractalkine between good prognosis group[(2.63±4.72)ng/mL]and poor prognosis group[(3.93±9.38)ng/mL](P>0.05).Conclusions Serum Fractalkine level is significantly decreased in patients with early-stage Parkinson′s disease,and the expression of serum Fractalkine may be closely related to the severity of Parkinson′s disease.Serum Fractalkine level of Parkinson′s disease patients has no significant guiding significance for prognosis evaluation.
