阿魏酸通过NLRP3炎症小体抑制辐射引起的小胶质细胞炎症 被引量:5

Ferulic Acid Inhibits Radiation-Induced Microglial Inflammation through NLRP3 inflammasome

摘要 目的:本文旨在探讨阿魏酸(Ferulic acid,FA)在辐射诱导的小胶质细胞炎症中的保护作用及其抗炎机制。方法:以8 Gy 60Co-γ射线建立BV-2小胶质细胞辐射炎症模型,采用细胞免疫荧光实验检测小胶质细胞IBA-1、NLRP3表达情况,ELISA实验检测小胶质细胞IL-1β分泌量的改变,Western blot实验检测小胶质细胞NLRP3、Caspase-1、IL-1β蛋白表达水平的变化。结果:与空白对照组相比,模型对照组小胶质细胞IBA-1、NLRP3表达增强(P<0.05),促炎因子IL-1β的分泌量明显增加(P<0.05),NLRP3、Caspase-1、IL-1β蛋白表达明显上调(P<0.05);与模型对照组相比,以2.5、5、10μmol/L阿魏酸预处理小胶质细胞,能够明显抑制辐射后小胶质细胞IBA-1、NLRP3表达的增强(P<0.05),显著减少细胞IL-1β的分泌(P<0.05),并使小胶质细胞中炎症调控因子NLRP3、Caspase-1和促炎因子IL-1β的蛋白表达水平明显下调(P<0.05)。结论:阿魏酸可能通过抑制NLRP3炎症小体的活化缓解辐射诱导的小胶质细胞炎症反应。 Objective:The purpose of this study was to investigate the protective effect and its mechanism of Ferulic acid(FA)on radiation-induced microglial inflammation.Methods:The radiation inflammation model of BV-2 microglia was established by 8 Gy 60Co-γray.The expression of Iba-1 and NLRP3 in microglia was detected by Immunofluorescence assay.The secretion of IL-1βin microglia was detected by ELISA assay.The protein expressions of NLRP3,Caspase-1 and IL-1βin microglia were detected by Western blot assay.Results:Compared with the control group,the expressions of Iba-1 and NLRP3 in microglia were significantly increased(P<0.05),and the secretion of inflammatory factor IL-1βin radiation model group was significantly increased(P<0.05),the protein expressions of NLRP3、Caspase-1 and IL-1βwere significantly up regulated(P<0.05).Compared with the radiation model group,the pretreatment of microglia at the doses of 2.5,5,10μmol/L FA inhibited the increased expression of Iba-1 and NLRP3 in microglia,they significantly reduced the secretion of IL-1βand down regulated the protein expressions of NLRP3,Caspase-1 and IL-1βin microglia(P<0.05).Conclusion:The results show that FA can inhibit the activation of microglia induced by radiation,its mechanism may be related to the inhibition of the activation of NLRP3 inflammasome.
