灯盏细辛胶囊对D-半乳糖诱导的模型小鼠认知障碍的改善 被引量:3

The Improvement Effect of Dengzhan Xixin Capsule on Cognitive Impairment of D-galactose-induced Mice

摘要 目的:研究灯盏细辛胶囊对D-半乳糖诱导的模型小鼠认知障碍的改善作用。方法:取健康KM小鼠颈背部皮下连续注射D-半乳糖125 mg/kg 4 w完成造模后,随机分为模型对照组、灯盏细辛200、400 mg/kg组和吡拉西坦500 mg/kg组,每组12只,连续7 w灌胃相应药液同时注射D-半乳糖,另设空白对照组。采用疲劳转棒仪测定小鼠从转棒上掉落次数和最大停留时间;采用开场实验记录小鼠直立数、静止时间、水平运动距离与运动轨迹;采用水迷宫实验记录小鼠逃避潜伏期、穿越平台次数;酶法检测血清中的超氧化物歧化酶(SOD)活力、丙二醛(MDA)、谷胱甘肽(GSH)含量以及海马组织中的乙酰胆碱(ACh)、乙酰胆碱酯酶(AChE)含量。结果:与空白对照组相比,模型对照组小鼠的掉落次数、静止时间、潜伏期均明显增加,最大停留时间、直立数、运动距离及穿越平台次数明显减少(P<0.05或P<0.01);同时SOD活力、GSH、ACh含量显著下降,MDA含量、AChE活力显著升高(P<0.01)。与模型对照组比较,灯盏细辛200、400 mg/kg组小鼠给药后第4、6、7 w掉落次数明显减少,最大停留时间增加,第2、3 d的静止时间明显减少,直立数与水平活动距离显著增加,灯盏细辛400 mg/kg组小鼠的逃避潜伏期明显缩短,穿越平台次数明显增加(P<0.05);灯盏细辛200、400 mg/kg组小鼠SOD活力、GSH、ACh含量明显升高(P<0.05或P<0.01),MDA含量、AChE活力显著降低(P<0.01)。结论:灯盏细辛胶囊能增强模型小鼠的学习记忆与情绪管理能力,提高其脑内胆碱水平,并对抗氧化损伤,对D-半乳糖致衰老小鼠认知功能障碍具有改善作用。 Objective:To study the effect of Dengzhan Xixin Capsule(EBC)on improving cognitive impairment in D-galactose induced mice.Methods:Healthy KM mice were subcutaneously injected with D-galactose(125 mg/kg)into the backs of the necks for 4 weeks to establish the model.Model mice were randomly divided into the model group,400 mg/kg EBC group,200 mg/kg EBC group and 500 mg/kg Piracetam group,12 mice per group,corresponding drugs were orally administered and D-galactose was injected simultaneously for 7 weeks,and the control group was also set.The fatigue rotating rod instrument was used to determine the number of dropping from rotating rod and the maximum residence time.The Open field test was used to record the number of mice standing upright,resting time,horizontal movement distance and movement trajectory.Morris water maze test was used to record the escaped latency and the number of crossing platforms.Then the enzyme method was used to detect serum superoxide dismutase(SOD),malondialdehyde(MDA),glutathione(GSH)and acetylcholine(ACh)cholinesterase(AChE)content in the hippocampal tissue.Results:Compared with the control group,the number of dropping,resting time,and latency time were increased in the model group,the maximum residence time,the number of standing up,the distance of movement and the number of crossing platforms were decreased(P<0.05 or P<0.01).the levels of SOD,GSH and ACh were significantly decreased,while the contents of MDA and AChE were significantly increased(P<0.01).Compared with the model group,in 400 mg/kg and 200 mg/kg EBC groups,the number of drops in the 4 th,6 th,and 7 th weeks after administration were significantly reduced,the maximum residence time was increased.On the 2 nd and 3 rd days,the resting time was reduced,the number of standing up and the distance of horizontal activity were significantly increased.The escape latency of mice in 400 mg/kg EBC group was significantly shortened,and the number of crossing platforms was significantly increased(P<0.05).At the same time,the activity of SOD and the contents of GSH and Ach were increased in 200、400 mg/kg EBC groups(P<0.05 or P<0.01),while the MDA content and AChE activity were significantly decreased(P<0.01).Conclusion:Dengzhan Xixin capsule can improve the learning,memory and emotion management abilities of model mice,resist oxidative stress damage,increase the choline level in the brain tissue,and improve the cognitive impairment of D-galactose induced aging mice.
