检测角度对激光诱导荧光光谱技术检测花生油AFB_(1)污染的影响研究

摘要 近年来,近红外光谱、高光谱等快速无损检测技术,被广泛应用于农产品中黄曲霉毒素B_(1)(Aflatoxin B_(1),AFB_(1))的检测。但这些方法主要是根据黄曲霉改变样品本身的结构结合化学计量学方法间接检测样品中AFB_(1),无特异性。因此,本研究拟利用自主研发的激光诱导荧光(laser induce fluorescence,LIF)技术获取污染AFB_(1)花生油的荧光光谱,建立精确无损检测花生油中AFB_(1)的定性、定量方法。首先,人工制备7种不同AFB_(1)污染水平的花生油样品,同时设置对照组。其次利用研发的LIF系统从不同角度检测样品的荧光光谱信号。最后,对获取的光谱进行分析。结果显示,三个检测角度的荧光强度在400~800 nm内均随着AFB_(1)污染水平的增加而增加,定性和定量的结果均是当检测荧光角度为90°时表现最优。表明当检测角度为90度时,LIF技术可以作为一种快速、精确的方法来判定花生油中AFB_(1)污染水平。 Recently,near infrared spectroscopy(NIRS),hyperspectral imaging(HSI)and other rapid non-destructive detection techniques have been shown potential for evaluation of aflatoxin B_(1)(AFB_(1))in agricultural products.However,these methods were mainly based on aflatoxin to change the structure of the sample itself.Then,no specificity was shown when combined with chemometrics to detect AFB_(1) in samples indirectly.Therefore,it was used in this study to develop laser induced fluorescence(LIF)technology and obtain the fluorescence spectrum of AFB_(1) contaminated peanut oil,thereby establishing an accurate and non-destructive qualitative and quantitative method for the detection of AFB_(1) in peanut oil.Firstly,seven kinds of peanut oil with different AFB_(1) pollution levels were prepared,and the control group was set up simultaneously.Secondly,the LIF system was adopted to detect the fluorescence spectrum signals of the samples from different angles.Finally,the obtained spectra were analyzed.The results showed that the fluorescence intensity of the three detection angles increased with the increase of AFB_(1) contamination levels in the range of 400~800 nm.Qualitative and quantitative results showed that the best performance was obtained when the fluorescence angle was 90°.LIF technology can be used as a rapid and accurate method to determine the level of AFB_(1) pollution in peanut oil.
