摘要
目的研究链霉菌C-1027对肝癌BEL-7402细胞的抗肿瘤作用,并探究其对BEL-7402细胞自噬的影响。方法采用MTT法检测C-1027对BEL-7402细胞活力的影响,台盼蓝排斥实验检测C-1027对BEL-7402细胞存活率的影响,克隆形成实验评价C-1027对BEL-7402细胞增殖的作用,AO染色检测自噬体的形成。结果C-1027处理剂量依赖性地降低了BEL-7402细胞的细胞活力,抑制BEL-7402细胞克隆形成。C-1027诱导BEL-7402细胞出现酸性自噬泡,呈剂量依赖性。结论C-1027具有明显的抗肿瘤活性,并且C-1027诱导BEL-7402细胞发生自噬。
Objective To examine the anti-tumor effect of streptomyces C-1027 on hepatocellular carcinoma BEL-7402 cells,and explore its effect on autophagy of BEL-7402 cells.Methods The MTT assay was used to detect the effect of C-1027 on cell viability of BEL-7402 cells.Trypan blue staining was used to detect the effect of C-1027 on the survival rate of BEL-7402 cells.The colony formation assay was used to assess the effect of C-1027 on cell proliferation of BEL-7402 cells.AO staining was used to detect the formation of autophagosomes.Results C-1027 treatment dose-dependently reduced cell viability of BEL-7402 cells,and the proliferation of BEL-7402 cells was significantly inhibited.Lidamycin induced acidic autophagic vesicles in BEL-7402 cells in a dose-dependent manner.Conclusion C-1027 exerts a significant anti-tumor effect,and C-1027 induces autophagy in BEL-7402 cells.
