摘要
目的检测环状RNA circ-STK39在肾癌中的表达,探讨其可能的调控通路。方法定量实时聚合酶链反应(qRT-PCR)检测31例肾癌及癌旁正常组织中circ-STK39的表达差异;应用RNase R核酸外切酶处理、Sanger测序验证circ-STK39的环化结构及稳定性;绘制接受者操作特征(ROC)曲线初步探讨circ-STK39作为肾癌分子标志物的潜在价值;生物信息学分析circ-STK39的基因特征,并预测其可能的调控通路。结果qRT-PCR结果显示circ-STK39在肾癌中较癌旁组织的表达显著上调;circ-STK39能耐受RNase R核酸外切酶,表明其环化结构的存在及稳定性;ROC曲线提示circ-STK39对肾癌具有一定的诊断价值,曲线下面积为0.734(95%CI:0.610~0.857),敏感性和特异性分别为0.871和0.516;生物信息学分析显示circ-STK39存在miR-192-5p的潜在结合位点,以此预测并构建了circRNA/miRNA/mRNA调控网络。结论circ-STK39在肾癌中高表达,其环化结构稳定,circ-STK39可能与miR-192-5p的调控有关。
Objective To detect the expression of circ-STK39 in renal cell carcinoma(RCC)and to explore the possible regulatory pathway.Methods The expression of circ-STK39 in 31 pairs of RCC and normal tissues adjacent to the cancer was detected with quantitative real-time polymerase chain reaction(qRT-PCR).The circular structure and stability of circ-STK39 were verified with RNase R treatment and Sanger sequencing.The receiver operating characteristic(ROC)curve was plotted to explore the potential value of circ-STK39 as a molecular marker for RCC.The genetic characteristics of circ-STK39 was analyzed and the possible regulatory pathway was predicted.Results The qRT-PCR results showed that the expression of circ-STK39 was significantly up-regulated in RCC compared with that in adjacent normal renal tissues.circ-STK39 tolerated RNase R exonuclease,indicating the existence and stability of its covalently closed loop.ROC curve indicated circ-STK39 had a potential diagnostic value for RCC.The area under the curve was 0.734(95%CI:0.610-0.857),and the sensitivity and specificity were 0.871 and 0.516,respectively.Bioinformatic analysis showed that circ-STK39 had a potential binding site for miR-192-5p.The circRNA/miRNA/mRNA regulatory network was predicted and constructed.Conclusion circ-STK39 is up-regulated in RCC,and its circular structure is stable.circ-STK39 may be related to the regulation of miR-192-5p in RCC.
