宫颈癌中Hippo-Yes相关蛋白1、雷帕霉素靶蛋白的表达及临床意义

摘要 目的探讨Hippo-Yes相关蛋白1(YAP1)、雷帕霉素靶蛋白(mTOR)在宫颈癌组织中的表达及意义。方法92例宫颈癌患者作为宫颈癌组,50例宫颈上皮内瘤变(CIN)作为CIN组,因子宫肌瘤行子宫切除的宫颈正常女性50例作为对照组,免疫组化法测定宫颈组织YAP1、mTOR蛋白表达,分析其与宫颈癌病理特点的关系。结果CIN组、宫颈癌组宫颈组织YAP1、mTOR蛋白表达阳性率高于对照组(P<0.05),宫颈癌组YAP1、mTOR蛋白表达阳性率高于CIN组(P<0.05);临床分期为Ⅲ期、分化程度为低分化、伴宫旁侵犯、脉管侵犯及淋巴结转移的宫颈癌患者宫颈组织YAP1、mTOR蛋白表达阳性率高于临床分期为Ⅰ~Ⅱ期、病理分化程度为中高分化、不伴宫旁侵犯、不伴脉管侵犯及无淋巴结转移的宫颈癌患者(P<0.05),宫颈浸润深度>1/3的宫颈癌患者mTOR阳性率高于宫颈浸润深度≤1/3的宫颈癌患者(P<0.05);宫颈癌癌组织YAP1、mTOR表达呈正相关(r=0.489,P<0.05)。结论宫颈癌患者癌组织YAP1、mTOR蛋白呈高表达,且与肿瘤分期、分化程度、肿瘤侵犯范围及淋巴结转移有关,两者可能协同影响宫颈癌细胞增殖及凋亡调节平衡,诱导宫颈癌发生及进展。 Objective To explore the expression and significance of Hippo-Yes associated protein 1(YAP1)and mammalian targetprotein of rapamycin(mTOR)in cervical cancer tissues.Methods Ninety-two patients with cervical cancer were collected as cervical cancer group.Fifty patients with cervical intraepithelial neoplasia(CIN)were collected as CIN group.Fifty patients with normal cervix who underwent hysterectomy for uterine fibroids were collected as control group.The expression of YAP1 and mTOR protein in cervical tissues were measured by using immunohistochemistry.Their relationship with the pathological characteristics of cervical cancer was analyzed.Results The positive rate of YAP1 and mTOR protein expression in cervical tissues in the CIN group and the cervical cancer group were higher than those in the control group(P<0.05),and the positive rate of YAP1 and mTOR protein expression in the cervical cancer group was higher than that in the CIN group(P<0.05).The positive rate of YAP1 and mTOR protein expression in cervical tissues of cervical cancer patients with clinical stage III,poorly differentiation,parauterine invasion,vascular invasion and lymph node metastasis was higher than that of patients with clinical stage I~II,moderately and highly pathological differentiation and without parauterine invasion,vascular invasion andlymph node metastasis(P<0.05).The positive rate of mTOR in cervical cancer patients with cervical invasion depth>1/3 was higher than that in cervical cancer patients with cervical invasion depth≤1/3(P<0.05).The expression of YAP1 and mTOR in cervical cancer tissues was positively correlated(r=0.489,P<0.05).Conclusion YAP1 and mTOR protein are highly expressed in cancer tissues of patients with cervical cancer The expression of YAP1 and mTOR proteinis related to tumor stage,degree of differentiation,tumor invasion,and lymph node metastasis.The two molecules may synergistically affect the balance of cervical cancer cell proliferation and apoptosis,and induce the occurrence and progression of cervical cancer.
