摘要
目的研究CD47在胃癌组织免疫微环境中的表达和浸润CD68^(+)TAMs(CD68 positive tumor-associated macrophages)和CD163^(+)TAMs(CD163 positive tumor-associated macrophages)的分布特征。方法采用免疫组化法检测CD47、CD68、CD163在78例胃癌组织样本中的表达,观察CD47的表达,计算肿瘤微环境中浸润CD68^(+)TAMs和CD163^(+)TAMs的分级,采用配对卡方检验进行统计分析。结果免疫组化染色显示CD47呈质膜型表达,在胃癌细胞和胃癌间质细胞中的表达明显强于其在癌旁胃黏膜腺体上皮细胞和间质细胞中的表达水平(P<0.05),且与淋巴结转移密切相关;同时,胃癌组织中浸润CD68^(+)TAMs和CD163^(+)TAMs的分级显著高于癌旁组织(P<0.01),但CD163/CD68比值在胃癌组织与癌旁组织中比较无差异。结论胃癌组织中CD47的高表达和CD68^(+)TAMs和CD163^(+)TAMs的浸润是胃癌的共性免疫微环境特征。
Objective To research the expression of CD47 and the infiltration of CD68 positive tumor-associated macrophages(CD68^(+)TAMs)and CD163 positive tumor-associated macrophages(CD163^(+)TAMs)in the gastric cancer immune microenvironment.Methods CD47 and TAMs(CD68 and CD163)expression were evaluated in 78 samples of gastric cancer tissues by immunohistochemistry.The expression of CD47 and the distribution of CD68^(+)TAMs and CD163^(+)TAMs in gastric cancer tissues were examined under microscope.The data was analyzed by Chi-square test.Results The results showed that CD47 was expressed in plasma membrane type,and the level of CD47 in gastric cancer cells and stromal cells was significantly higher than that in adjacent gastric tissues(P<0.05),and it was closely related to lymph node metastasis.Meanwhile,the levels of CD68^(+)TAMs and CD163^(+)TAMs in gastric cancer microenvironment were significantly higher than those in paracancerous tissues(P<0.01).However,there was no difference in CD163^(+)/CD68^(+)ratio between gastric cancer tissues and paracancerous tissues.Conclusion The high expression of CD47 in gastric cancer tissues and the infiltration of CD68^(+)TAMs and CD163^(+)TAMs in gastric cancer microenvironment may be the common significant immune microenvironment characteristics.
