The role of SHCBP1 in the development of non-small cell lung cancer

摘要 目的:探讨纺锤体相关蛋白1(SHCBP1)在非小细胞肺癌(NSCLC)发生发展中的作用。方法:选取2018年1月到2020年3月在赣南医学院第一附属医院治疗的NSCLC患者62例,通过RT-PCR、Werstern blotting和免疫组化法检测SHCBP1基因在NSCLC中的表达并检测其蛋白的表达与定位;利用试剂盒构建SHCBP1质粒并合成出具有特异性的siRNA及shRNA,将之转染至NSCLC细胞株,利用RT-PCR和Western blotting对mRNA及其蛋白表达量进行检测,通过CCK-8实验以及克隆形成检测SHCBP1对NSCLC增殖的影响;利用划痕实验检测过表达及SHCBP1对NSCLC细胞迁移能力的影响。结果:在所检测的样本中,NSCLC组织中SHCBP1的表达相比较非癌组织更高(P<0.01);SHCBP1在NSCLC组织中主要表达于细胞的细胞质中。过表达研究发现SHCBP1能对NSCLC的增殖及迁移起到促进作用,对SHCBP1进行干扰能够抑制增殖和迁移。结论:SHCBP1在NSCLC中能够促进细胞增殖和迁移,对其发生过程有促进作用。 Objective: To explore the role of SHCBP1 in the development of non-small cell lung cancer( NSCLC). Methods: Select 62 patients with NSCLC treated in the first affiliated hospital of Gannan medical university from January 2018 to March 2020,and detected the expression of SHCBP1 gene in NSCLC by RT-PCR,Werstern blotting and immunohistochemistry and detected its protein expression and localization of SHCBP1;used the kit to construct SHCBP1 plasmid and synthesized specific siRNA and shRNA to transfect it into non-small cell lung cancer cell lines,used RT-PCR and Western blotting to detect mRNA and its protein expression,through CCK8 and clone formation to detect the effect of SHCBP1 on the proliferation of NSCLC;used the scratch test to detect the effect of overexpression and SHCBP1 on the migration ability of NSCLC cells. Results: Among the samples tested,the expression of SHCBP1 in NSCLC tissue was higher than that in non-cancer tissue( P < 0. 01);the expression of SHCBP1 in NSCLC was mainly found in the cytoplasm of cells. After overexpression studies,it was found that SHCBP1 can promote the proliferation and migration of NSCLC,and interference with SHCBP1 can inhibit proliferation and migration. Conclusion: SHCBP1 can promote cell proliferation and migration in non-small cell lung cancer,and has a role in promoting its occurrence.
