摘要
目的研究玛咖对虚寒、虚热证大鼠环核苷酸、内分泌激素及物质代谢水平的影响,验证玛咖偏温中药药性。方法SD大鼠分为空白组、虚寒模型组、虚寒肉桂组、虚寒玛咖高剂量组、虚寒玛咖低剂量组、虚热模型组、虚热黄柏组、虚热玛咖高剂量组、虚热玛咖低剂量组;采用每日肌肉注射氢化可的松琥珀酸钠、地塞米松磷酸钠注射液1次,连续21天的方法制备虚寒、虚热证动物模型;通过放免法检测环磷酸腺苷(cyclic adenosine monophosphate,cAMP)、环磷酸鸟苷(cyclic guanosine monophosphate,cGMP)、三碘甲状腺原氨酸(three iodine thyroid,T3)、四碘甲状腺原氨酸(thyroxine,T4)、反三碘甲状腺原氨酸(reverse triiodothyronine,rT3)、皮质酮(corticosterone,CORT)、睾酮(testosterone,T)、雌二醇(estradiol,E2)含量及总胆固醇(total cholesterol,TC)、甘油三酯(triglyceride,TG)、游离脂肪酸(free fat acid,FFA)、脂蛋白脂肪酶(lipoprteinlipase,LPL)、总蛋白(total protein,TP)、白蛋白(albumin,ALB)和血糖(glucose,GLU)含量变化。结果(1)玛咖高剂量能明显升高虚寒证大鼠血清cAMP、cAMP/cGMP、T3、T4、CORT、E2、TC、TG、FFA、LPL、TP、ALB、GLU值(P<0.05),降低rT3和T的含量(P<0.05);(2)玛咖低剂量可以明显升高虚寒证大鼠血清cAMP/cGMP、T3、T4、CORT、E2、TC、TG、FFA、TP、ALB、GLU值(P<0.05);(3)玛咖高剂量和玛咖低剂量均能明显降低虚热证大鼠血清E2、TC、TG、FFA值(P<0.05),升高CORT、T含量(P<0.05);(4)玛咖对虚热证大鼠cAMP、cAMP/cGMP、T3、T4、rT3、TP、GLU、LPL指标的影响无统计学意义(P>0.05)。结论本实验采用同类比较、异类反证的方法,通过测定玛咖对虚寒、虚热证模型大鼠环核苷酸、内分泌激素及脂肪、蛋白质和糖代谢等相关指标的含量变化,证明了玛咖偏温药性。
Objective To study the effects of Maca on the cyclic nucleotide,hormones and metabolism levels in rats with deficiency-cold pattern and deficiency-heat pattern,and to explore the Chinese medicine properties of Maca.Methods SD rats were divided into blank group,deficiency-cold pattern group,deficiency-cold Cinnamomi Cortex group,high and low-dose Maca groups,deficiency-heat pattern group,deficiency-heat Phellodendri Chinensis Cortex group,and high and low-dose Maca groups.The rats were injected hydrocortisone 21 days to set up the deficiency-cold model,and injected dexamethasone sodium phosphate to set up the deficiency-heat model.Detecting the contents of cAMP,cGMP,T3,T4,rT3,CORT,T,E2,TC,TG,FFA,LPL,TP,ALB,GLU by radioimmunoassay.Results(1)In Maca high-dose group,cAMP,cAMP/cGMP,T3,T4,CORT,E2,TC,TG,FFA,LPL,TP,ALB,GLU values were significantly increased(P<0.05),and the content of T and rT3 decreased(P<0.05).(2)In Maca low-dose group,cAMP/cGMP,T3,T4,CORT,E2,TC,TG,FFA,TP,ALB,GLU values were significantly increased(P<0.05).(3)Compared with the deficiency-heat model group,both the Maca high-dose group and low-dose group can increase the contents of CORT,T(P<0.05),decrease the values of E2,TC,TG,and FFA,ALB(P<0.05).(4)But the contents of cAMP,cAMP/cGMP,T3 and T4 had no statistical significance.Conclusion In this study,the content changes of cyclic nucleotides,endocrine hormones,fat,protein,and sugar metabolism related indicators were preliminarily determined to prove that Maca’s medicinal properties is slightly warm by the methods of homogeneous comparison and heteroge-neous disproval.
