摘要
目的通过采用网络药理学方法,探讨参芪地黄汤治疗糖尿病肾病(DN)的作用机制.方法利用TCMSP数据库平台查找参芪地黄汤的有效化学成分及与化学成分相关的潜在靶点,并在UniProt数据库中对靶点的名称标准化.再利用Gene Cards数据库、OMIM数据库筛选出与DN相关的疾病靶点,并通过Cytoscape 3.7.2软件构建成分-靶点-基因-疾病之间相互作用的网络关系图,进而在STRING数据库中绘制蛋白-蛋白PPI网络图,采用Rx643.6.2软件及Bioconductor生物信息软件包对药物与疾病的交集基因做GO功能富集和KEGG通路分析.结果筛选出参芪地黄汤的50个活性成分可能通过作用于AGE-RAGE、MAPK、HIF-1、IL-17等167条信号通路,来参与调节PGR、NOS2、PTGS1、AR、PTGS2等133个DN疾病的靶点,从而发挥治疗DN的作用.结论通过本次网络药理学的研究初步揭示了参芪地黄汤多成分、多靶点、多通路治疗DN的作用机制,为临床和科研提供依据和参考.
Objective To explore the mechanism of Shenqidihuang Decoction in the treatment of diabetic nephropathy based on network pharmacology.Methods The(TCMSP)database plat-form was used to find the effective components and potential targets which related to the ef-fective components of Shenqidihuang Decoction.The names of the targets were standardized in the UniProt database.The Gene Cards database and OMIM database was used to screen out disease targets which related to diabetic nephropathy,following by a network diagram of com-ponent-target-gene-disease interactions were constructed by Cytoscape 3.7.2.The protein-protein PPI network in the STRING database was drawn.Rx643.6.2 software and Bioconductor biological information software package were used to analyze GO function enrichment and KEGG pathway on the intersection genes of drugs and diseases.Results The 50 active compo-nents of Shenqidhuang decoction may affect 137 significant signaling pathways including AGERAGE,MAPK,HIF-1,and IL-17 by regulating 133 diabetic nephropathy disease targets such as PGR,NOS2,PTGS1,AR and PTGS2,so as to play a role in the treatment of diabetic nephropa-thy.Conclusion Through this network pharmacology study,the mechanism of Shenqi Dihuang Decoction's multi-component,multi-target,and multi-channel treatment of diabetic nephropa-thy was initially revealed,which provided basis and reference for clinical and scientific research.
