摘要
目的采用科学、全面的实验设计方法,确定川贝母水提液的最佳制备工艺。方法实验设计使用中心组合设计响应面法(RSCCD),优化考察提取时间、液料比和提取次数等参数;评价指标为水提液中西贝母碱含量,使用高效液相色谱法测定,色谱柱为Waters BEH C 18(柱长25 cm,直径0.46 cm,粒径5μm),检测器为蒸发光散射检测器,流动相为乙腈-0.02%二乙胺水溶液(25∶75,v/v),流速1.0 mL·min^(-1),漂移管温度为40℃,氮气流速2.4 mL·min^(-1)(增益1),柱温25℃,进样量10μL;根据Design Expert 8.02软件计算结果得出最优工艺参数。结果当提取条件为煎煮55 min,液料比1∶25(g∶mL),提取两次时,所得水提液中西贝母碱含量最高,为2.416 mg·mL^(-1)。结论本研究对川贝母水提液的制备工艺进行了科学全面的优化,所得结果符合预期。
OBJECTIVE To confirm the optimum preparation technique for Fritillaria cirrhosa bulbs decoction,by using scientific,comprehensive experimental design.METHODS The experiment design used response surface central composite design(RSCCD),parameters like extraction time,solvent-material ratio and extraction cycle were evaluated and optimized;Sipeimine content in the obtained decoction were used as index,the determination performed on high performance liquid chromatography(HPLC),column used was Waters BEH C 18(length 25 cm,inner diameter 0.46 cm,particle size 5μm),the detector was evaporative light scattering detector(ELSD),the mobile phase was acetonitrile-0.02%diethylamine solution(25∶75,v/v),the flow rate was 1.0 mL·min^(-1),the drift tube temperature was 40℃,nitrogen flow rate was 2.4 mL·min^(-1)(gain 1),column temperature was 25℃,the injection volume was 10μL;optimum technique was calculated by Design Expert 8.02 software.RESULTS When the extraction conditions were set as follows:extraction time 55 min,solvent-material ratio 1∶25(g∶mL),extraction cycle 2,the obtained decoction had highest Sipeimine content,the value was 2.416 mg·mL^(-1).CONCLUSION Our study scientifically and comprehensively optimized the preparation technique for Fritillaria cirrhosa bulbs decoction,which was in line with our forcast.
