槐生多年卧孔菌净油抗氧化和抗肿瘤活性评价与化学成分分析

摘要 目的评价药用真菌槐生多年卧孔菌净油抗氧化和抗肿瘤活性,并分析其化学成分。方法用乙醚提取槐生多年卧孔菌净油,通过1,1-二苯基-2-苦基肼基(DPPH)清除试验评价其抗氧化活性,采用噻唑蓝(MTT)法评价其抗肿瘤活性,运用气相色谱串联质谱(GC-MS)分析净油化学成分。结果生物活性试验结果表明,槐生多年卧孔菌净油有一定的抗氧化活性,清除DPPH自由基的IC_(50)值为2.71 mg·mL^(-1),但对受试肿瘤细胞(Hela、A549和B16F10)抑制活性不明显,IC_(50)值均大于100μg·mL^(-1)。通过GC-MS鉴定了槐生多年卧孔菌净油中的12种成分,占净油总量的43.47%。结论槐生多年卧孔菌净油有一定的抗氧化活性,而且抗氧化活性随净油浓度的增大而增强,但抗肿瘤活性不明显。大环麝香类化合物黄葵内酯、1-氧杂环十四烷-2-酮和2-羟基环十五烷酮是槐生多年卧孔菌净油具有独特浓郁优雅麝香香气的主要原因。 OBJECTIVE To evaluate the antioxidant and antitumor activities of the absolute oil from the medicinal mushroom Perenniporia robiniophila(Murrill)Rvarden,and to analysis its chemical composition.METHODS The absolute oil was isolated from Perenniporia robiniophila(Murrill)Rvarden by ether,and the antioxidant activity was evaluated by DPPH free radical scavenging test,and the antitumor activities were evaluated by MTT method.It′s chemical composition were analyzed by GC-MS.RESULTS The biological activity test showed that the absolute oil had a certain antioxidant activity,with the IC_(50) value of 2.71 mg·mL^(-1) against DPPH radical scavenging,but the inhibitory activities against the tested tumor cell lines(Hela,A549 and B16F10 cell lines)were not obvious,all with the IC_(50) values of>100μg·mL^(-1).12 components in the absolute oil were identified,accounting for 43.47%of the total amount of the absolute oil.CONCLUSION The absolute oil of Perenniporia robiniophila(Murrill)Rvarden had a certain antioxidant activity,and the antioxidant activity increased with the increase of concentration of the absolute oil,but its antitumor activities were not obvious.The macrocyclic musks,such as ambrettolid,1-oxazacyclotetradecane-2-one and 2-hydroxycyclopentadecanone,are the main reasons for the unique rich and elegant musk fragrance of the absolute oil.
