辣椒素通过PI3K/AKT信号通路对鼻咽癌细胞增殖、凋亡及侵袭的抑制作用 被引量:3

Capsaicin inhibits proliferation,apoptosis and invasion of nasopharyngeal carcinoma cells through PI3K/AKT signaling pathway

摘要 目的探讨辣椒素通过PI3K/AKT信号通路对鼻咽癌细胞增殖、凋亡及侵袭能力抑制作用的影响。方法选择120只SPF级BALB/c小鼠,用鼻咽癌细胞株建立鼠荷瘤模型,建模成功后根据处理方式不同分空白对照组(不做任何处理)、低剂量实验组(加入5.0μmol/L辣椒素)及高剂量实验组(加入7.5μmol/L辣椒素),采用四唑盐比色法检测鼻咽癌细胞增殖能力,流式细胞仪测定细胞凋亡率,Transwell法测定细胞侵袭能力,Western blot法测定抑制PI3K/AKT信号通路后p-AKT、AKT、BCL-2、MMP-2、MMP-9蛋白表达。结果细胞转染培养24 h、48 h后,3组细胞吸光度值均明显升高(P <0.05)。细胞转染培养24 h后,高剂量实验组鼻咽癌细胞凋亡率高于低剂量实验组,低剂量实验组低于空白对照组(P <0.05)。高剂量实验组p-AKT、AKT、BCL-2、MMP-2、MMP-9蛋白表达低于低剂量实验组,低剂量实验组低于空白对照组,差异有统计学意义(P <0.05)。结论辣椒素通过调节PI3K/ATK信号通路对鼻咽癌组织细胞增殖、侵袭能力进行抑制,加快肿瘤细胞凋亡,提高患者预后生存质量。 Objective To explore the effect of capsaicin on the inhibition of proliferation,apoptosis and invasion of nasopharyngeal carcinoma cells through PI3 K/AKT signaling pathway. Methods A total of 120 cases of SPF BALB/c mice were selected to establish a mouse tumor-bearing model with nasopharyngeal carcinoma cell lines. After successful modeling,they were divided into blank control group( no treatment) and low-dose experimental group( adding 5. 0 μmol/L capsaicin) and the high-dose experimental group( adding 7. 5 μmol/L capsaicin). Tetrazolium salt colorimetric method was used to detect the proliferation ability of nasopharyngeal carcinoma cells,the cell apoptosis rate was measured by flow cytometry,the cell invasion ability was measured by the Transwell method,and the p-AKT,AKT,BCL-2,MMP-2,MMP-9 protein expression after inhibiting the PI3 K/AKT signaling pathway were measured by Western-blot. Results After the cells were transfected and cultured for 24 h and 48 h,the absorbance values of the three groups of cells all increased significantly( P < 0. 05). After cell transfection and culture for 24 hours,the apoptosis rate of nasopharyngeal carcinoma cells in the high-dose experimental group was higher than that in the low-dose experimental group,and the low-dose experimental group was lower than the blank control group( P < 0. 05). The protein expression of p-AKT,AKT,BCL-2,MMP-2,and MMP-9 in the high-dose experimental group was lower than that in the low-dose experimental group,and the low-dose experimental group was lower than the blank control group,with the differences statistical significance( P < 0. 05). Conclusion Capsaicin inhibits the proliferation and invasion of nasopharyngeal carcinoma cells by regulating the PI3 K/ATK signaling pathway,accelerates tumor cell apoptosis,and improves the prognostic quality of life of patients.
