基于NLRP3炎症小体探讨天麻钩藤颗粒对自发性高血压肝阳上亢证认知障碍的影响 被引量:3

摘要 目的:探讨天麻钩藤颗粒对自发性高血压肝阳上亢证认知功能障碍的改善及血清炎症因子NLRP3、ASC、Caspase-1的影响。方法:将32只自发性高血压大鼠,随机选8只作为自发性高血压组,其余大鼠连续28 d给予附子汤(2 g·kg^(-1))复制自发性高血压肝阳上亢证大鼠模型,然后将其分为自发性高血压肝阳上亢证组、天麻钩藤颗粒组(3.125 g·kg^(-1))、尼莫地平片组(6.25 g·kg^(-1)),另设6只SD大鼠为空白组,连续给予相应的药物42 d。观察大鼠的一般情况;Morris水迷宫检测大鼠穿越平台次数;ELISA法检测血清NLRP3、ASC、Caspase-1水平;HE染色观察大鼠脑组织病理损伤变化。结果:与自发性高血压肝阳上亢组比较,天麻钩藤颗粒组大鼠逃避潜伏期缩短,目标象限停留时间延长,穿越平台次数增多(P<0.05);大鼠收缩压、舒张压均显著降低(P<0.01);可显著降低大鼠血清NLRP3、Caspase-1、ASC水平(P<0.01),同时大鼠额叶病理损伤有所改善。结论:天麻钩藤颗粒能够抑制NLRP3炎症小体合成,保护脑额叶神经元,改善大鼠的认知能力。 Objective: To study the effect of Tianma Gouteng Granule on cognitive impairment and serum inflammatory factors NLRP3,ASC and caspase-1 in spontaneous hypertension with hyperactivity of liver yang syndrome.Methods: 32 spontaneously hypertensive rats were randomly selected as the spontaneously hypertensive group.The other rats were given Fuzi Decoction(2 g·kg^(-1)) for 28 days to replicate the spontaneously hypertensive rat model with hyperactivity of liver yang syndrome.Then they were divided into spontaneous hypertension with hyperactivity of liver yang syndrome group, Tianma Gouteng granule group(3.125 g·kg^(-1)) and nimodipine tablet group(6.25 g·kg^(-1)).Another 6 SD rats were set as the normal group, and the corresponding drugs were given continuously for 42 days.The general condition of rats was observed;Morris water maze was used to detect the number of rats crossing the platform;the levels of serum NLRP3,ASC and caspase-1 were detected by ELISA;HE staining was used to observe the pathological changes of brain tissue in rats.Results: Compared with the spontaneously hypertensive group with hyperactivity of liver yang syndrome, the escape latency of rats in Tianma Gouteng granule group was shortened, the residence time in target quadrant was prolonged, and the number of crossing platform was increased(P<0.05);Systolic blood pressure and diastolic blood pressure decreased significantly(P<0.01);it can significantly reduce the levels of serum NLRP3,ASC and caspase-1(P<0.01),and improve the pathological injury of frontal lobe.Conclusion: Tianma Gouteng granule can inhibit the synthesis of NLRP3 inflammatory body, protect frontal lobe neurons and improve the cognitive ability of model rats.
