补肾疏肝法联合五行音乐对肝郁型小鼠卵巢储备功能的影响

Effect of Bushen Shugan Method Combined with Five Elements Music on Ovarian Reserve Function in Liver-Suppressed Mice

目的:探讨补肾疏肝法联合五行音乐对肝郁型小鼠卵巢储备功能的影响。方法:将健康清洁级雌性KM小鼠随机分为4组,即空白组、模型组、补肾疏肝方联合音乐组(26 g·kg^(-1)+音乐刺激)和逍遥散组(28 g·kg^(-1)),每组10只。除空白组外,其他小鼠采用改良慢性不可预见性应激法建立肝郁型小鼠模型。模型成功后灌胃给药,空白组和模型组给予等体积的生理盐水灌胃,每日1次,连续21 d。实验开始后,每日观察小鼠的一般情况;小鼠阴道脱落细胞涂片判定小鼠的动情次数;糖水偏嗜度实验检测小鼠的糖水偏好;旷场实验考察小鼠在中央格的停留时间;ELASA法检测小鼠血清中雌二醇(estradiol, E2)、促卵泡刺激素(lutropin alfa hormone, FSH)的水平;HE染色观察小鼠卵巢病理变化。结果:与空白组比较,模型组小鼠体质量、动情次数、糖水偏嗜度及E2的水平明显降低(P<0.05),中央格停留时间明显延长(P<0.05),FSH的水平明显升高(P<0.05),卵巢结构明显破坏;与模型组比较,补肾疏肝方联合音乐组小鼠体质量、动情次数、糖水偏嗜度及E2的水平显著升高(P<0.05),中央格停留时间明显缩短(P<0.05),卵巢病理情况有所改善。结论:肝郁状态下小鼠卵巢储备功能降低,补肾疏肝法联合五行音乐对肝郁型小鼠卵巢功能的恢复有一定的促进作用。

Objective: To investigate the effect of Bushen Shugan Method combined with Five Elements Music on ovarian reserve function in liver-suppressed mice.Methods: Healthy and clean female KM mice were randomly divided into 4 groups: blank group, model group, Bushen Shugan Prescription combined with music group(26 g·kg^(-1)+music stimulation) and Xiaoyao Powder group(28 g·kg^(-1)),with 10 mice in each group.In addition to the blank group, other mice used the improved chronic unpredictable mild stress method to establish the mouse model of liver depression.After the success of the model, the blank group and the model group were given the same volume of normal saline once a day for 21 days.After the experiment, the general conditions of mice were observed every day;the number of estrus in mice was determined by vaginal exfoliated cell smear;the preference of mice for sugar water was detected by sugar water preference test;the residence time of mice in the central grid was investigated by open field experiment.The levels of estrodiol(E2) and lutropin alfa hormone(FSH) in serum of mice were detected by ELASA method;HE staining was used to observe the pathological changes of mouse ovary.Results: Compared with the blank group, the body weight, estrous times, sugar water preference and E2 level in the model group decreased significantly(P<0.05),the central grid residence time prolonged significantly(P<0.05),the level of FSH increased significantly(P<0.05),and the ovarian structure was damaged significantly;compared with the model group, the body weight, estrous times, sugar water preference and E2 level of mice in the Bushen Shugan Prescription combined with music group were significantly increased(P<0.05),the central grid residence time was significantly shortened(P<0.05),and the ovarian pathology was improved.Conclusion: Under the condition of liver depression, the ovarian reserve function of mice decreased.The method of Bushen Shugan combined with Five Elements Music can promote the recovery of ovarian function in liver depression mice.
