摘要
在体外通过加入不同浓度的硝苯地平与日本血吸虫雄虫共培养,探讨硝苯地平杀伤成虫作用及相关机制。结果表明:0.05-1.00μg/mL硝苯地平作用下雄虫活力值降低,且虫体活力值与硝苯地平浓度之间呈负相关。硝苯地平预孵育可部分恢复吡喹酮作用后的日本血吸虫雄虫活力,但两者联合用药后虫体仍表现为痉挛、卷曲,皮层结构可见损伤变化。电压门控钙离子通道阻断剂硝苯地平体外对日本血吸虫成虫具有杀伤作用,可拮抗吡喹酮对日本血吸虫的杀伤作用。
S.japonicum male worms were collected for exploring the antischistosomal effect of voltage-gated calcium channels antagonist nifedipine in vitro cultivation,the results showed that there are statistically significant differences in the viability score of male worms exposed to nifedipine.There is a negative liner correlation between the viability scores of male worms and nifedipine concentrations in the range of 0.05-1.00μg/mL.Preincubation with nifedipine can partially restore the viability of S.japonicum males exposed to praziquantel,and the worms shows curved or spiral comma shaped,along with evident destruction of the tegument surface.In summary,nifedipine,a voltage-gated calcium channels antagonist,has been showed the antischistosomal effects on S.japonicum adult worm,but can affect the killing effect of praziquantel on the parasite.
