摘要
目的:探讨桃红化浊丸对四氯化碳(CCl_(4))致肝纤维化模型大鼠的影响。方法:采用腹腔注射体积分数40%CCl_(4)复制大鼠肝纤维化模型,将模型成功的大鼠随机分为模型组、秋水仙碱(0.5 mg·kg^(-1))组、桃红化浊丸(1.54 g·kg^(-1))组,另设空白组,每组10只,连续给予相应的药物6周。Western blot法检测大鼠肝脏Smad3和金属蛋白酶组织抑制因子-1(the tissue inhibitor of metalloproteinases-1,TIMP-1)蛋白表达,RT-PCR法检测大鼠肝脏miRNA29b、Smad3 mRNA和TIMP-1 mRNA水平。结果:与模型组比较,桃红化浊丸组能明显下调大鼠肝脏Smad3、TIMP-1蛋白表达,升高miRNA29b水平,降低Smad3 mRNA、TIMP-1 mRNA水平。结论:桃红化浊丸可能是通过增加肝脏下调大鼠肝脏Smad3、TIMP-1蛋白表达,升高miRNA29b水平,降低Smad3 mRNA、TIMP-1 mRNA水平,从而达到防治肝纤维化的作用。
Objective: To inquire into the influence of Peach Kernel Turbidity-Resolving Pill on rats with hepatic fibrosis induced by CCl_(4).Methods: The rat model of hepatic fibrosis was established by intraperitoneal injection of 40% CCl_(4).The modeled rats were randomly divided into the model group, the colchicine(0.5 mg·kg^(-1)) group and the group of Peach Kernel Turbidity-Resolving Pill(1.54 g·kg^(-1)).Another blank group was set, with 10 rats in each group, and the corresponding drugs were given continuously for 6 weeks.The expressions of Smad3 and TIMP-1 protein in rat liver were detected by Western blot, and the levels of miRNA29 b, Smad3 mRNA and TIMP-1 mRNA in rat liver were detected by RT-PCR.Results: Compared with the model group, the group of Peach Kernel Turbidity-Resolving Pill could significantly down-regulate the expressions of Smad3 and TIMP-1 protein, increase the level of miRNA29 b, and reduce the levels of Smad3 mRNA and TIMP-1 mRNA.Conclusion: Peach Kernel Turbidity-Resolving Pill may prevent and treat liver fibrosis by increasing the expression of Smad3 and TIMP-1 protein, increasing the level of miRNA29 b and reducing the levels of Smad3 mRNA and TIMP-1 mRNA.
