基于网络药理学探讨马齿苋药理作用的分子机制 被引量:2

摘要 目的:运用网络药理学的方法探讨马齿苋药理作用的分子机制。方法:以"马齿苋"为检索词,在中药系统药理学数据库与分析平台进行检索,根据口服生物利用度及类药性参数查找有效成分及其靶基因,构建有效成分-靶点基因网络作用图;构建蛋白质相互作用(protein-protein interation, PPI)网络图,筛选出相互作用的核心靶点并对其作用靶点进行GO富集分析和KEGG通路富集分析,明确马齿苋具体的作用分子机制。结果:共筛选出10种马齿苋的有效成分、221个靶点蛋白,靶蛋白PPI网络中其中的核心靶点蛋白包括AKT1、ALB、白细胞介素-6(interleukin-6,IL-6)等。富集分析发现马齿苋作用的靶点蛋白多分布于神经突触后膜、细胞质的核周区域及细胞外基质等区域、分子功能涉及蛋白质同源二聚体激活、转录因子结合、蛋白质结构域特异性结合等;生物过程富集于对有机环状化合物及有毒物质的反应等;生物通路包括多条癌症通路、乙型肝炎通路及动脉粥样硬化通路。结论:马齿苋在抗癌、心血管等方面有潜在的药理作用。 Objective: To inquire into the molecular mechanism of pharmacological action of Machixian(Herba Portulacae) based on network pharmacology.Methods: Machixian(Herba Portulacae) was used as a search term to search in the pharmacology database and analysis platform of traditional Chinese medicine system.According to oral bioavailability and drug-like parameters, the active components and their target genes were found, active component-target gene network action diagram was constructed.The protein-protein interaction(PPI) network diagram was constructed;the core targets of the interaction were screened, and the action targets were analyzed by GO enrichment and KEGG pathway enrichment, so as to clarify the specific molecular mechanism of Machixian.Results: A total of 10 active components and 221 target proteins of Machixian were screened.The core target proteins in the PPI network of target proteins included AKT1,ALB,interleukin-6(IL-6),etc.Enrichment analysis showed that the target proteins of Machixian were mostly distributed in the postsynaptic membrane, perinuclear region of cytoplasm and extracellular matrix.The molecular functions involved protein homodimer activation, transcription factor binding, protein domain specific binding and so on.Biological processes concentrated in reactions to organic cyclic compounds and toxic substances.Biological pathways included multiple cancer pathways, hepatitis B pathways and atherosclerosis pathways.Conclusion: Machixian has potential anti-cancer and pharmacological effects in cardiovascular and other aspects.
