香水莲花提取物对黄嘌呤氧化酶的抑制作用 被引量:4

摘要 目的:评价香水莲花对黄嘌呤氧化酶(XOD)的抑制效果,进一步分析抑制类型。方法:以别嘌醇为阳性对照,利用紫外分光光度法检测单位时间内酶促反应体系中尿酸含量的变化,计算出各抑制剂的抑制率以及半数抑制浓度,根据酶动力学分析香水莲花对XOD的抑制类型、抑制动力学常数Ki。结果:香水莲花水提物对XOD有一定的抑制作用,抑制类型为可逆的竞争性抑制,抑制动力学常数Ki为2.01 mg/mL。4个地区样本中,广东中山水提物抑制效果最好,IC50为64.72μg/mL。结论:香水莲花水提物中含有抑制XOD的有效成分,作为XOD抑制剂的开发具有一定的优势。 Objective:The inhibitory effect of Nymphaeahybrid on Xanthine oxidase(XOD)was proved and the type of inhibition was further analyzed.Methods:Allopurinol was used as positive control.Ultravi⁃olet spectrophotometry was used to detect the change of uric acid content in enzymatic reaction system in unit time,and the inhibitory rate and half maximal inhibitory concentration of each inhibitor were calcu⁃lated.According to the analysis of enzyme kinetics,the type of inhibition of XOD and inhibit kinetic con⁃stant Ki by Nymphaeahybrid was analyzed.Results:The extract of Nymphaeahybrid had certain inhibitory effect on XOD.The inhibition of XOD by water extract was a reversible competitive inhibition and inhibit kinetic constant Ki was 2.01 mg/mL.Amongthe 4 regions,the best inhibition effect was found in Zhong⁃shan,Guangdong,and the half maximal inhibitory concentration was 64.72μg/ml.Conclusions:The ex⁃tracts of Nymphaeahybrid contain active components that inhibit XOD,and the development of Nympha⁃eahybridas XOD inhibitor has certain advantages.
