摘要 目的总结活化T细胞核因子5(NFAT5)对实体肿瘤的影响以及促进肿瘤发生发展的作用机制,分析NFAT5在肿瘤学领域中相关研究的不足及未来方向。方法应用PubMed以"NFAT5、TonEBP、OREBP、cancer、tumor"等为关键词检索文献,同时以"活化T细胞核因子5、癌"等为关键词检索中国期刊全文数据库。检索时间为1999-03―2020-10,共检索到英文相关文献195篇,中文文献39篇。纳入标准:(1)NFAT5相关调控因素;(2)NFAT5在实体瘤中的相关研究;(3)NFAT5对免疫微环境的影响。根据标准共纳入45篇文献。结果NFAT5对多种类型的实体肿瘤发挥着促癌作用。NFAT5既可以通过直接调控AQP5、S100A4等促进癌细胞的增殖、运动能力等,也可以通过增强其他转录因子的活性而发挥间接作用。结论 NFAT5在部分实体肿瘤的发展及转移中起着重要作用,针对NFAT5的治疗可能会改变肿瘤的免疫微环境,有望为这些肿瘤的治疗提供新的思路。 Objective To summarize the effects of NFAT5 on solid tumors and shed light on the mechanism, and analyz the shortcomings of precious studies and pointed out the future research direction.Methods "NFAT5,TonEBP,OREBP,cancer, tumor" were searched as keywords by PubMed.Meanwhile, related articles published on CNKI databases were searched with keywords such as "nuclear factor of activated T cell 5,cancer".The search time was from March 1999 to October 2020.Totally 195 related English articles and 39 Chinese articles were retrieved.Inclusion criteria:(1) regulatory factors for NFAT5;(2) effects of NFAT5 on solid tumors;(3) roles of NFAT5 in the immune microenvironments.A total of 45 articles were included according to the criteria.Results In most studies, NFAT5 shows cancer promoting effects in a variety types of solid tumors.NFAT5 can promote proliferation and motility of cancer cells by direct control of the downstream genes such as AQP5,S100 A4 and so on.Also, it can promote cancers as an enhanceosome in an indirect way.Conclusions NFAT5 plays important roles in the development and metastasis of some solid tumors.NFAT5 has special meanings for the immune microenvironment, which provides new strategies for tumor treatment.
