摘要
为催化卤化产物,挖掘具有生物活性的新卤化物提供新的方法途径,拟构建卤化酶原核表达载体催化天然卤化物的卤化过程。对链霉菌Streptomyces sp.FJS31-2基因组中一个II型PKS类产物zunyimycin生物合成基因簇进行系列分析,针对其中的后修饰酶卤化酶基因和单加氧酶基因进行原核表达纯化。通过分子生物学手段成功构建卤化酶基因(65 kD)和单加氧酶基因(37 kD)的原核表达载体,并对其进行鉴定。卤化酶和单加氧酶在催化化合物的卤化过程中,会产生疑似新卤化产物。
A new method for catalyzing halogenated products was mined new halides with biological activity,intending to construct a prokaryotic expression vector of halogenase to catalyze halogenation process of natural halides.A series of sequences analyses of a type II PKS product zunyimycin biosynthesis gene cluster in genome of Streptomyces sp.FJS31-2 was carried out,aiming at the prokaryotic expression and purification of post-modification enzyme halogenase gene and monooxygenase gene were carried out.The results showed that prokaryotic expression vector of the halogenase gene(65 kD)and monooxygenase gene(37 kD)was successfully constructed and identified by molecular biology means.It thus concludesd that alogenases and monooxygenases could produce ambiguous new halogenated products in the process of catalyzing the halogenation of compounds.
