北京市两个白纹伊蚊种群kdr抗性突变检测分析 被引量:6

Detection and analysis of kdr resistance mutations in two populations of Aedes albopictus in Beijing

摘要 目的对2个白纹伊蚊野外种群的kdr抗性突变进行检测分析,评估其对拟除虫菊酯类杀虫剂的抗性风险。方法利用PCR扩增白纹伊蚊钠离子通道基因(voltage-gated sodium channel, VGSC)的两个结构域并进行测序,分析与击倒抗性(knockdown resistance,kdr)有关位点(1 016、1 532和1 534位点)的突变与系统进化关系。结果在2个种群的60只白纹伊蚊中,检测到位点1 016发生突变的有3只,占5.00%;位点1 534发生突变的有2只,占3.33%;位点1 532发生突变的有17只,占28.33%。对应的序列单倍型分析显示种群内kdr突变具有多种进化起源。结论白纹伊蚊野外种群在拟除虫菊酯类杀虫剂选择压力下,其靶标发生了多位点kdr突变,今后伊蚊防治应加强环境治理和蚊幼控制,合理使用拟除虫菊酯类杀虫剂控制成蚊进而延缓抗性产生。 Objective To detect the kdr resistance mutations of two wild populations of Aedes albopictus and assess their risk of resistance to pyrethroid insecticides.Methods The two domains of the voltage-gated sodium channel(VGSC) of A.albopictus were amplified by PCR and sequenced to analyze the mutations at the sites(1 016,1 532 and 1 534) related to knockdown resistance(kdr),as well their correspondent haplotype phylogenetic relationship.Results The above three sites were detected in 60 specimen of two A.albopictus populations.Among them, the frequencies of mutations at site 1 016 and 1 534 were relatively lower(5.00% and 3.33%,respectively),and the one at site 1 532 was higher(28.33%);their sequence haplotype analysis showed that the kdr mutations in the populations had multiple evolutionary origins.Conclusion Under the long-term selection pressure of pyrethroid insecticides, the wild populations of A.albopictus showed multiple sites of kdr mutations in the target gene VGSC.The control of Aedes mosquitoes should be strengthened with environmental management and larvae control, and using sprayed-pyrethroids rationally in control adult mosquito in order to delay the development of resistance.
