摘要
目的探讨CLEC7A在颅内动脉瘤(IAA)组织和血清中的表达及临床意义。方法检索GEO数据库中IAA基因数据集,分析IAA基因数据集中CLEC7A的表达水平及其与患者年龄、性别的关系。收集IAA患者血清标本60例,并收集健康人血清标本20例作为对照,采用ELISA法检测血清中CLEC7A表达水平,分析IAA血清中CLEC7A表达水平与患者年龄、性别的关系,分析血清CLEC7A水平对IAA的诊断价值。结果两个IAA基因数据集(GSE75436和GSE54083)的分析结果显示,CLEC7A在IAA组织中的表达水平显著高于正常动脉组织(P<0.05);GSE54083数据集分析结果显示,不同年龄段和性别的IAA组织中CLEC7A表达水平差异无统计学意义(均P>0.05)。IAA患者血清中CLEC7A表达水平也显著高于健康人血清水平,且年龄高(≥60岁)的患者血清CLEC7A水平表达较年龄低(<60岁)的患者高(均P<0.05);血清CLEC7A在男性和女性患者的表达水平差异无统计学意义(P>0.05)。受试者工作特征(ROC)曲线显示,血清CLEC7A水平对IAA有一定的诊断价值(灵敏度为0.618,特异度为0.700)。结论CLEC7A在IAA组织和血清中的表达水平均较健康人显著升高,提示CLEC7A与IAA的发病密切相关,血清CLEC7A水平可作为IAA的辅助诊断指标之一。
Objective To investigate the expression and clinical significance of CLEC7A in intracranial aneurysms(IAA)tissues and serum.Methods IAA gene dataset was searched in the GEO database to analyze the expression of CLEC7A in the IAA gene dataset and its relationship with age,gender of patients.A total of 60 serum specimens from IAA patients were collected,and other 20 serum specimens from healthy people were also collected as controls,the ELISA method was employed to detect serum CLEC7A expression,the relationship between CLEC7A expression in IAA serum and age,gender of patients was analyzed,and the value of serum CLEC7A level on diagnosing IAA was also analyzed.Results The analysis results of two IAA gene dataset(GSE75436 and GSE54083)revealed that the expression of CLEC7A was significantly higher in IAA tissues than in normal arterial tissues(P<0.05).The analysis result of GSE54083 dataset interpreted that there were no statistically significant differences in CLEC7A expressions in IAA tissues of different age groups and genders(all P>0.05).The IAA patients obtained a significantly higher expression of serum CLEC7A as compared with the healthy people,and the people in high age(≥60 years of age)obtained a higher expression of serum CLEC7A in the comparison of the people in low age(<60 years of age)(all P<0.05).There was no statistically significant difference of serum CLEC7A expression between male and female patients(P>0.05).Receiver operating characteristic(ROC)curve revealed that serum CLEC7A level had certain diagnostic value on IAA(with sensitivity in 0.618,and specificity in 0.700).Conclusion Compared to the indicators mentioned below in the healthy people,expressions of CLEC7A more significantly increased in IAA tissues and serum,which present that CLEC7A is closely correlated to the morbidity of IAA,and serum CLEC7A level can be used as one of the auxiliary diagnostic indicator of IAA.
