摘要
目的:探讨腺苷酸环化酶相关蛋白2(CAP2)在结肠癌组织及癌旁组织中的表达及临床价值。方法:回顾性分析2018年10月-2020年10月本院收治的50例结肠癌患者的临床资料。取患者结肠癌组织及癌旁组织,采用实时荧光定量PCR(RT-qPCR)法测定组织CAP2 mRNA表达,采用免疫组化法测定组织CAP2蛋白表达,并测定术前血清癌胚抗原(CEA)、CA19-9水平。分析CAP2蛋白表达与临床病理特征相关性;采用Kendall相关分析CAP2蛋白与CEA、CA19-9的相关性;采用受试者工作特征(ROC)曲线评估CAP2对结肠癌的诊断价值。结果:结肠癌组织CAP2 mRNA表达水平明显高于癌旁组织(P<0.05)。结肠癌组织CAP2蛋白阳性表达率为70.00%,明显高于癌旁组织的16.00%(P<0.05)。不同分化程度、浸润程度、TNM分期、淋巴结转移、术前CEA及CA19-9水平的结肠癌组织CAP2蛋白阳性表达率比较,差异均有统计学意义(P<0.05)。Kendall相关分析显示,结肠癌组织中CAP2蛋白阳性表达率与术前血清CEA、CA19-9均呈正相关(τ=0.467、0.544,P<0.001)。ROC曲线显示,CAP2 mRNA、CAP2蛋白对结肠癌具有一定诊断价值(AUC=0.895、0.770,P<0.001)。结论:结肠癌组织CAP2表达水平明显高于癌旁组织,且其高表达与患者分化程度、浸润程度、TNM分期、淋巴结转移、术前CEA及CA19-9水平密切相关,提示CAP2参与了结肠癌发生、进展,可能成为治疗结肠癌潜在靶点。
Objective:To investigate the expression and clinical value of adenylate cyclase related protein 2 (CAP2) in colon cancer tissue and adjacent tissue.Method:The clinical data of 50 patients with colon cancer admitted to our hospital from October 2018 to October 2020 were retrospectively analyzed.The colon cancer tissues and adjacent tissues were taken,real-time fluorescence quantitative PCR (RT-qPCR) was used to determine the expression of CAP2 mRNA in tissues,and CAP2 protein in tissues by immunohistochemistry.Serum carcinoembryonic antigen (CEA) and CA19-9 levels were measured before operation.The correlation between CAP2 protein expression and clinicopathologic features was analyzed.Kendall correlation was used to analyze the correlation between CAP2 protein and CEA,CA19-9.The diagnostic value of CAP2 in colon cancer was evaluated by receiver operating characteristic (ROC) curve.Result:The expression level of CAP2 mRNA in colon cancer tissues was significantly higher than that in adjacent tissues (P<0.05).The positive expression rate of CAP2 protein in colon cancer tissues was 70.00%,which was significantly higher than 16.00% in adjacent tissues (P<0.05).There were statistically significant differences in the positive expression rate of CAP2 protein in colon cancer tissues with different degrees of differentiation,degree of invasion,TNM stage,lymph node metastasis,preoperative CEA and CA19-9 level (P<0.05).Kendall correlation analysis showed that the positive expression rate of CAP2 protein in colon cancer tissue was positively correlated with serum CEA and CA19-9 before operation (τ=0.467,0.544,P<0.001).ROC curve showed that CAP2 mRNA and CAP2 protein had certain diagnostic value in colon cancer (AUC=0.895,0.770,P<0.001).Conclusion:The expression level of CAP2 in colon cancer tissues is significantly higher than that in adjacent tissues,and its high expression is closely related to the degree of differentiation,degree of invasion,TNM stage,lymph node metastasis,preoperative CEA and CA19-9 levels,suggesting that CAP2 is involved in the occurrence and progression of colon cancer,and may become a potential target for the treatment of colon cancer.
