电针刺激对外伤性脊髓损伤大鼠脊髓CaMKⅡ表达及神经功能恢复的影响 被引量:3

Effect of electroacupuncture on the expression of CaMKⅡand the recovery of nerve function in the spinal cord of traumatic spinal cord injury of rats

摘要 目的观察电针刺激足阳明胃经穴位对外伤性脊髓损伤(TSCI)大鼠受损脊髓Ca MKII mRNA及蛋白表达、大鼠神经功能恢复的影响。方法将96只SD大鼠按照不作电针干预、作督脉穴位电针刺激、作足阳明胃经穴位电针刺激3种干预方式平均分为损伤对照组、电针对照组、胃经电针组,每组平均分为1、2、3、4周亚组,钳夹法复制TSCI大鼠模型,对比各组各时间点BBB功能评分、采用qRT-PCR及Western blot方法对比各组Ca MKII mRNA及蛋白表达水平。结果与损伤对照组相比,电针对照组及胃经电针组同时段的BBB评分显著升高(P<0.05),前3周胃经电针组显著高于同时段电针对照组;与损伤对照组相比,胃经电针组同时段的Ca MKII mRNA及蛋白均显著降低(P<0.05),损伤对照组及电针对照组、电针对照组及胃经电针组均无显著差别(P>0.05)。结论电针刺激足阳明胃经可通过抑制受损脊髓节段Ca MKII mRNA及蛋白的表达从而改善TSCI大鼠的神经功能,且依据不同中医理论进行取穴,在分子生物学机制及疗效上,存在差异。 Objective To observe the effect of electroacupuncture on the expression of Ca MKII mRNA and protein and the recovery of nerve function in the injured spinal cord of rats with TSCI.Methods 96 SD rats were divided into injury control group,electroacupuncture control group and stomach meridian electroacupuncture group.According to the three intervention methods,no electroacupuncture intervention,electric acupuncture stimulation of Du Meridian and Foot Yangming stomach meridian acupoint electroacupuncture stimulation.Each group was divided into 1,2,3 and 4 week subgroups.TSCI rat model was reproduced by clamp method.Compare BBB function scores of each time point in each group.The expression of Ca MKII mRNA and protein was compared by QRT PCR and Western blot.Results Compared with the injury control group,BBB score of electroacupuncture control group and electroacupuncture through stomach group in the same period was significantly higher(P<0.05),In the first three weeks,the electroacupuncture group was significantly higher than the electroacupuncture control;Compared with the control group,the expression of Ca MKII mRNA and protein in the same period of electroacupuncture group was significantly lower(P<0.05).There was no significant difference between injury control group and electroacupuncture control group(P>0.05).There was no significant difference between electroacupuncture control group and electroacupuncture group(P>0.05).Conclusion-Electroacupuncture stimulation of Foot Yangming stomach meridian can improve the neurological function of TSCI rats by inhibiting the expression of Ca MKII mRNA and protein in injured spinal cord segments.There are differences in molecular biological mechanism and curative effect of acupoint selection according to different TCM theories.
